Differing performance of two assays for the measurement of anti‐Mullerian hormone in premenopausal women: A cross‐sectional study

Abstract: Objective To compare the performance of two anti‐Mullerian (AMH) assays over a range of concentrations, in samples collected from young women. Design A cross‐sectional method‐comparison study of 168 non‐healthcare‐seeking women. Participants Included women were aged 18–39 years, not recently pregnant, breast feeding or using systemic hormones. Measurements Serum AMH levels were analysed with the Beckman Coulter Access 2 assay from fresh samples and the Ansh picoAMH assay using samples stored at −80°C, in a par… Show more

“…These include the use of multiple different assays from a variety of manufacturers, reflecting the evolution of AMH measurement across nearly two decades of research. These assays differ in their sensitivities, limits of detection, and limits of quantification and are not interchangeable ( Iliodromiti et al , 2017 ; Bungum et al , 2018 ; Bell et al , 2021b ). This limited direct between-study comparisons of the number of women with undetectable AMH as well as comparisons of prognostic models.…”

“…This difference may in part be due to the use of older, less sensitive assays in previous studies, which can result in higher values at lower concentrations. 34 We also compared patterns of AMH over time with those in the published literature. A study that measured AMH in healthy term girls (n = 13 girls) at 2 days and 1 month of age showed an increase in AMH over this time period.…”

“…These kits, i.e., ultrasensitive AMH and picoAMH ELISA, can detect very low blood AMH levels. Although these kits use the same antibodies and calibrators, picoAMH ELISA can cover the lower range of the standard curve 73–78 . Good correlations of assay values have been found between these new kits and conventional kits 79–82 (Table 1).…”
