Direct hapten-linked multiplexed immunoassays on polycarbonate surface

Abstract: Direct hapten-linked multiplexed immunoassay is developed on the polycarbonate surface of standard Digital Versatile Discs (DVDs) for six compounds of environmental concern, as proof of concept. Carboxylated haptens are directly linked to the aminated polycarbonate surface through carbodiimide/succinimide coupling. The modified DVD surface maintained its physical and optical properties. Multiplexed assay reached detection limits down to 0.1 μg/L for chlorpyrifos, 2,4,5-trichlorophenoxypropionic acid, sulfathia… Show more

“…For CD/DVD based assays, the protocol should not affect either the surface or optical path of the detection system. Several groups have demonstrated methods using chemically functionalized polycarbonate surfaces of conventional CDs or DVDs to enable biomolecules binding, 26,27 where the formation of -COOH groups on the PC surface was achieved by UV. However, limited success has been achieved mainly because of the low surface density of reactive groups and strong non-specific adsorption.…”

“…In addition, several attempts have been made to use optical scanning of spinning disks as the basis for immunoassays. A group in Madrid has been developing technology based on standard disks (33)(34)(35)(36)(37)(38). In the most recent version (38 ), 6 environmentally related small molecules are determined in a competition assay with haptens directly linked to the plastic surface, and colloidal gold antibody is used for label with silver intensification.…”

“…Considerable effort has thus devoted to developing fast and accurate approaches for malathion analyses. Enzyme-linked immunosorbent assays (ELISA) (Brun et al, 2005;Tamarit-López et al, 2011) represent one of the most typical and powerful strategies among them. In ELISA, antibodies are indispensable for analyte recognition and subsequent signal reporting in ELISA for malathion detection.…”
