Direct Infusion MS-Based Lipid Profiling Reveals the Pharmacological Effects of Compound K-Reinforced Ginsenosides in High-Fat Diet Induced Obese Mice

Abstract: The serum lipid metabolites of lean and obese mice fed normal or high-fat diets were analyzed via direct infusion nanoelectrospray-ion trap mass spectrometry followed by multivariate analysis. In addition, lipidomic biomarkers responsible for the pharmacological effects of compound K-reinforced ginsenosides (CK), thus the CK fraction, were evaluated in mice fed high-fat diets. The obese and lean groups were clearly discriminated upon principal component analysis (PCA) and partial least-squares discriminant ana… Show more

“…Lipid profiling was performed using Thermo LTQ XL ion trap MS (Thermo Fisher Scientific, West Palm Beach, FL, USA) as previously described by Shon et al . (). A detailed information of lipid metabolites with MS/MS fragmentation were already constructed and provided in LipidBlast libraries (Kind et al ., ).…”

“…Each dried extract was reconstituted in 100 μL of solvent mixture (methanol/chloroform, 9:1, v/v) and diluted 50-fold with methanol/chloroform (9/1, v/v) containing 7.5 mM ammonium acetate. Lipid profiling was performed using Thermo LTQ XL ion trap MS (Thermo Fisher Scientific, West Palm Beach, FL, USA) as previously described by Shon et al (2015). A detailed information of lipid metabolites with MS/MS fragmentation were already constructed and provided in LipidBlast libraries (Kind et al, 2013).…”

“…The resulting data matrix was processed using SIMCA-P+ (version 12.0, Umetrics, Umea, Sweden) for multivariate statistical analysis. For data processing of lipid profiles, nominal ion mass spectra data files from the ion trap mass spectrometer (“.raw” files) were directly loaded into the Genedata Expressionist MSX module (Genedata AG, Basel, Switzerland) [ 25 ]. Data on all detected peaks, including m / z and intensity values, were exported as Excel files.…”

“…These results are in line with a previous study showing that treatment of HFD-fed mice with compound K-enriched with ginsenosides, which decreases hepatic steatosis in treated mice, also reduced serum levels of TAG species with a total number of carbons above 50. 56 Remarkably, in the serum of patients with obesity and NAFLD, similar TAG species (TAG (C50:1/2/3) and TAG (C52:3/4)) are reduced 6 months after bariatric surgery. These results are in line with previous studies reporting a reduction in TAG (C50:1), among other TAGs, in plasma/ serum of obese patients after RYGB interventions.…”
