Direct preconditioning of cultured chick ventricular myocytes. Novel functions of cardiac adenosine A2a and A3 receptors.

Abstract: Preconditioning with brief ischemia before a sustained period of ischemia reduces infarct size in the perfused heart. A cultured chick ventricular myocyte model was developed to investigate the role of adenosine receptor subtypes in cardiac preconditioning. Brief hypoxic exposure, termed preconditioning hypoxia, prior to prolonged hypoxia, protected myocytes against injury induced by the prolonged hypoxia.

“…A protective e ect following A 3 receptor stimulation at the cardiac level has been described in di erent animal models (Strickler et al, 1996;Stambaugh et al, 1997;Tracey et al, 1997) but the lack of any signi®cant e ect of the A 3 selective agonist IB-MECA (Gallo-Rodriguez et al, 1994) suggests that an A 3 -mediated in¯uence of adenosine on cardiac function can be excluded.…”

“…At the cardiac level, A 3 receptor stimulants, administered prior to and during an ischaemic episode, have shown a protective action on heart cells in human and other animals (Strickler et al, 1996;Stambaugh et al, 1997;Tracey et al, 1997;Carr et al, 1997). Finally, a decrease in BP without any change in HR, mediated by A 3 receptor stimulation was observed in the rat by Stella et al (1998).…”

“…Acting through the A 2A adenosine receptor (AR), it suppresses prolonged inflammation [2] and causes vasodilation and inhibits platelet aggregation, thus increasing the amount of oxygen available to an organ under stress. Adenosine agonists selective for the A 3 AR are of interest as cerebroprotective [3], cardioprotective [4,5], and anticancer [6] agents.…”

“…A 3 agonists are also protective in the heart, as well as the brain. In a chick ventricular myocyte culture (8) and in the isolated rabbit heart (9) activation of A 3 receptors by IB-MECA induced preconditioning, which greatly improved the outcome following an ischemic injury.…”
