Direct vaccination with tumor cells killed with ICP4-deficient HSVd120 elicits effective antitumor immunity

Abstract: We tested whether tumor cells were killed by replication-incompetent recombinant herpes simplex virus (HSV) d120 lacking immediate early gene ICP4 and whether HSVd120-killed tumor cells could be used directly for tumor vaccination. Vaccine efficacy was tested in TC-1, a murine adenocarcinoma transformed with HPV16 E6 and E7, and ID8-Vegf, a murine epithelial ovarian cancer model. HSVd120 killed tumor cells by apoptosis. Tumor cells infected by HSVd120 were engulfed more avidly by immature DCs and induced DC ma… Show more

“…A vaccine composed of a nonreplicating HSV infected tumor that is irradiated and then presented to dendritic cells has been shown to produce a strong T cell response including the stimulation of tumor reactive IFN- γ secreting T cells and tumor reactive cytotoxic T cells [25]. IL-12 boosts the Th1 immune response and stimulates the production of IFN- γ and TNF- α .…”

“…The mice were euthanized whenever the tumor ulcerated or tumors exceeded a size of 15 mm in any of the perpendicular diameters. The tumor volume (V) was determined by using the formula, V=1/2(Length) (Width) 2 [26,27]. …”
