Discovery by serendipity: a new context for an old riddle

Abstract: Abstract. In the last years there has been a great improvement in the development of computational methods for combinatorial chemistry applied to drug discovery. This approach to drug discovery is sometimes called a "rational way" to manage a well known phenomenon in chemistry: serendipity discoveries. Traditionally, serendipity discoveries are understood as accidental findings made when the discoverer is in quest for something else. This 'traditional' pattern of serendipity appears to be a good characterizati… Show more

“…27 In some cases, an individual may be searching for a new idea, problem, solution, or opportunity. In combinatorial chemistry, for instance, which often yields new drugs, the notion of "a blind search" is part of the process, 28 with "serendipity mistakes" just a likely stage in the experiment. Going down a blind alley in search of some answer but finding another one, then, is almost built into the research process itself.…”

“…Serendipity has been recognized as an important component in many fields, such as scientific research [9], art [22] and humanistic research [20]. The main point of study, especially in creative thinking, has been the strive for understanding how different serendipitous encounters take place [7].…”

“…Our ability to design target chemical species or synthetic routes (usually via target-oriented synthesis, TOS) is predicated on previous knowledge, including models that attempt to explain patterns in chemical reactivity. New data is key to the development of materials, methods, or models, and serendipity often plays a large part in this process. , Methods that “engineer” serendipity into the chemical discovery process are of interest but hard to achieve when developing reliable approaches for library generation, for example. Despite this, library generation is a powerful tool for gathering relevant data for pattern finding. It is possible to classify libraries as either “Focused” or “Prospecting,” depending on the region of chemical space accessed by members.…”
