Dissecting the Underlying Pharmaceutical Mechanism of Chinese Traditional Medicine Yun-Pi-Yi-Shen-Tong-Du-Tang Acting on Ankylosing Spondylitis through Systems Biology Approaches

Abstract: Traditional Chinese Medicine (TCM) has been served as complementary medicine for Ankylosing Spondylitis (AS) treatment for a long time. Yun-Pi-Yi-Shen-Tong-Du-Tang (Y-Y-T) is a novel empirical formula designed by Prof. Chengping Wen. In this study, a retrospective investigation supported efficacy of Y-Y-T and then we deciphered the underlying molecular mechanism of the efficacy. Herbal ingredients and targeting proteins were collected from TCMID. PPI networks were constructed to further infer the relationship … Show more

“…Ankylosing spondylitis falls into the categories of “nephroparalysis” and “bone impediment” in traditional Chinese medicine (TCM) [ 8 – 10 ]. In the treatment of ankylosing spondylitis, Chinese medicine mainly focuses on replenishing the liver and kidneys, replenishing qi and blood, dispelling rheumatism, and relieving arthralgia [ 11 , 12 ]. The clinical application of TCM in the treatment of ankylosing spondylitis is based on a wealth of experience.…”

“…Network pharmacology is an approach to predict disease-specific targets from biomedical data available in systems biology and polypharmacology [ 19 ]. With the development of biomedical data, new methods based on “active ingredient-target-disease” and interaction networks have been developed to understand the complex pharmacological mechanisms of Chinese medicine, and have blossomed in recent years [ 20 , 21 ]. Network pharmacology's fundamental principles and the holistic view of Chinese medicine are intertwined, leading to a change from the traditional “one target, one drug” paradigm to the more recent “multicomponent, multitarget” model, which is the most effective model for addressing multitarget medications [ 22 , 23 ].…”

“…The study of AS in Traditional Chinese Medicine (TCM) has a long history and has accumulated extensive experience in treating AS, and some Chinese herbal formulas (CHF) have been confirmed to have better clinical effects on this disease [22] . On the basis of the holistic concept and syndrome differentiation, TCM can be directed at different pathogenic links, using a variety of methods, and through multiple pathways, to achieve the effect of treating diseases and regulating immunity, and can alleviate the side effects of western medicine [23] . Therefore, integrated TCM and western medicine in the treatment of AS can synergize with each other to exert their own advantages, which can both remove multiple pathogenic factors of AS and improve the systemic status to achieve the best therapeutic effect [24] .…”
