Distinct Peripheral Blood RNA Responses to Salmonella in Pigs Differing in Salmonella Shedding Levels: Intersection of IFNG, TLR and miRNA Pathways

Abstract: Transcriptomic analysis of the response to bacterial pathogens has been reported for several species, yet few studies have investigated the transcriptional differences in whole blood in subjects that differ in their disease response phenotypes. Salmonella species infect many vertebrate species, and pigs colonized with Salmonella enterica serovar Typhimurium (ST) are usually asymptomatic, making detection of these Salmonella-carrier pigs difficult. The variable fecal shedding of Salmonella is an important cause… Show more

“…[96,97]), while studies in the pig or cow disproportionately revolve around resistance to infection, in particular Mycobacterium bovis in cattle and Salmonella in pigs (e.g. [98][99][100]). Reproductive biology studies are also well represented (e.g.…”

“…Six piglets (one from each sow) were housed separately from ST-inoculated piglets, and were inoculated with PBS alone, as described previously. 15 Data were collected at 0, 1, 2, 7, 14 and 21 DPI. Bacteriology for qualitative and quantitative analysis of Salmonella from rectal swabs was performed as described previously.…”

“…Further, we have previously determined that there is little difference in gene expression in peripheral blood sampled at 20 DPI for LS and PS pigs. 15 RNA was purified by DNase I digestion and the RNeasy Mini Elute Cleanup Kit as recommended by the manufacturer (Qiagen). RNA samples were confirmed to be DNA-free by lack of genomic DNA on PCR amplification.…”

“…2 of 18 In addition, TLRs also react to damage associated molecular patterns (DAMPs) [2,3] released after cellular damage, and thus play a crucial role in initiating the innate immune response [4]. Genetic variations found in the genes encoding TLRs have been associated with disease susceptibility and resistance in a variety of animal species [1,5,6]. In the bovine genome, 10 members of the TLR family (TLR1-10) have been identified and mapped to specific chromosomes [7].…”
