Distinction between breast cancer cell subtypes using third harmonic generation microscopy

Abstract: Third Harmonic Generation (THG) microscopy as a non-invasive, label free imaging methodology, allows linkage of lipid profiles with various breast cancer cells. The collected THG signal arise mostly from the lipid droplets and the membrane lipid bilayer. Quantification of THG signal can accurately distinguish HER2-positive cells. Further analysis using Fourier transform infrared (FTIR) spectra reveals cancer-specific profiles, correlating lipid raft-corresponding spectra to THG signal, associating thus THG to … Show more

“…Non-linear microscopy (NLM) diagnostic modalities are also included in Table 1. Non-linear microscopy techniques (NLM), such as MPEF, second harmonic generation (SHG), and third harmonic generation (THG), are well-established techniques for biological research [109][110][111][112][113][114]. These techniques under different modalities provide new insights for the assessment of appropriate conservation methods that have to be applied for various CH objects of high artistic or cultural significance [79,115].…”

“…SHG enables the detection of high degree-organized structures without inversion symmetry, such as stacked membranes and arranged proteins (e.g., collagen) [80,111]. THG allows the resolution of transparent interfaces, on the basis of local differences in refractive index and dispersion [79,110,112].…”

“…More important, THG microscopy has shown great potential for clinical applications. Excellent agreement with the standard histopathology of skin cancers has been demonstrated for THG [10,11] and THG also shows great potential for breast tumor diagnosis [12,13].…”

“…More studies are needed to improve the therapeutic interventions for the treatment of human breast cancers [12][13][14]. Different subtypes of breast-cancer cells vary with respect to prognosis, aggressiveness, and response to therapies.…”
