Dithiol-PEG-PDLLA Micelles: Preparation and Evaluation as Potential Topical Ocular Delivery Vehicle

Abstract: Thiol-modified nanoparticles have potential applications in mucoadhesive drug delivery and have been examined in this regard for topical ocular delivery. In this paper we provide a simple method for the synthesis of a dithiol terminated amphiphilic diblock copolymer. Bidentate dithiol-poly(ethylene glycol)-poly(d,l-lactide) (SH2-PEG-PDLLA) was synthesized and micelles with dithiol-containing coronas were prepared from this block copolymer via the emulsion method. In vitro release studies indicated that the pre… Show more

“…Instead, the product mixture was directly used for the subsequent polymerization step, since HOOC-PEG-COOH was inert under the ring-open polymerization conditions. As a result, a monocarboxyl-terminated block-polymer HOOC-PEG-PLA was obtained in the presence of unreacted HOOC-PEG-COOH, which was able to be removed by hot water washing [25]. Finally, HEMI was connected to HOOC-PEG-PLA via an esterification reaction, affording Mal-PEG-PLA.…”

“…5 Mucoadhesivity of micelles can be improved by incorporating the functional groups, which are capable of binding to the chemical groups present within mucosa. 77 Thiol is a good example of these functional groups that can interact with cysteine that is available in abundance in the mucin layer. 78 Therefore, materials containing thiol groups can be easily attached to the mucin‏ layer and thus enhance the residence time.…”

“…In comparison to our systems, other polymeric micelles recently used for Dex encapsulation exhibited lower loading capacities. A loading capacity in the range 0.5–3% was observed for Dex-loaded micelles composed of poly(ethylene glycol) (PEG) as a hydrophilic part, and poly(butyl methacrylate) 53 , poly(D,L-lactide) 54 , poly(Δ-caprolactone) (PCL) 55 , 56 and poly(propylene glycol) 57 , respectively, as a hydrophobic core. Slightly higher loading capacities (from 7 to 12%) were reported for PEG-PCL micelles loaded with Dex-acetate.…”
