Diverse Molecular Targets for Chalcones with Varied Bioactivities

Abstract: Natural or synthetic chalcones with different substituents have revealed a variety of biological activities that may benefit human health. The underlying mechanisms of action, particularly with respect to the direct cellular targets and the modes of interaction with the targets, have not been rigorously characterized, which imposes challenges to structure-guided rational development of therapeutic agents or chemical probes with acceptable target-selectivity profile. This review summarizes literature evidence o… Show more

“…Chalcones possess a wide range of biological activities, mainly against parasites [7 -9] and, several antiparasitic mechanisms have been reported and proposed. [12,21] Considering that chalcones 1 and 3 causing extensive morphological alteration on the tegument of adult schistosomes, ATPDase inhibition may not be the only suggested target of schistosomicidal action and other mechanisms may be involved. In this regard, it is reported that the therapeutic potential of chalcones is related to their structure as α,β-unsaturated ketones and, therefore, to their action as Michael acceptors, which could capture nucleophiles, such as cysteine, and bind covalently to proteins and enzymes.…”

“…In many instances, a single chalcone may demonstrate multiple bioactive properties. These diverse bioactivities may be attributed to the flexible structural conformation of the chalcone backbone, leading to promiscuous substrate behavior [11]. Two chalcones, 4-hydroxyderricin (1) and xanthoangelol (2), are the most abundant in this plant and possess cytotoxic, anti-inflammatory, and antidiabetic properties [12].…”
