DNA Associated with Circulating Exosomes as a Biomarker for Glioma

Abstract: Cancerous and non-cancerous cells secrete exosomes, a type of nanovesicle known to carry the molecular signature of the parent for intercellular communications. Exosomes secreted by tumor cells carry abnormal DNA, RNA, and protein molecules that reflect the cancerous status. DNA is the master molecule that ultimately affects the function of RNA and proteins. Aberrations in DNA can potentially lead a cell to malignancy. Deviant quantities and the differential sequences of exosomal DNA are useful characteristics… Show more

“…KRAS mutation is also confirmed in exosomes of pancreatic ductal adenocarcinoma in the early stage and the late phase ( Allenson et al, 2017 ). The serum exosomes of glioma patients also carry the gDNA sequence with the same biological mutation as glioblastoma ( Wang et al, 2018 ; Vaidya and Sugaya, 2020 ). In addition, the corresponding driving mutation was also found in the exosomes of prostate cancer ( Lazaro-Ibanez et al, 2014 ).…”

“…Exosomes also contain DNA molecules representing introns, non-coding genomic DNA, and other chromosomal sequences, including transposable elements. Several mutated genes specific to brain glioma have been found in exosomes, such as MGMT [22][23][24], PTEN [22], EGFR [25,26], and many others [27]. We aimed to verify whether the exoDNA concentration in the plasma of patients affected by glioma was related to the volume and histological grading of their tumors.…”

“…Tumor biomarker is a term used to describe a potential indicator of cancer development and progression specific to a particular cancer type, like proteins, nucleic acids (DNA, mRNA, miRNA), and EVs [ 25 , 34 ]. Ideally, a new biomarker should fulfill an unmet need in cancer detection; alternatively, it should provide advantages over preexisting biomarker(s), such as being more accurate and simpler to measure, faster, or cheaper [ 35 ].…”
