DNA Double-strand Break Induction in Yeast by X-rays and α-particles Measured by Pulsed-field Gel Electrophoresis

Abstract: Pulsed-field gel electrophoresis was used to separate the chromosomes of the diploid yeast Saccharomyces cerevisiae 211*B after irradiation with X-rays and alpha-particles. After electrophoresis, gels were stained with ethidium bromide, placed on a UV-transilluminator and photographed with a digitizing camera connected to a personal computer. The pictures obtained were processed with the help of specially developed software which allows for the correction of the camera's shading effect and background fluoresce… Show more

“…Experimental details of both methods have been published (Akpa et al 1992;L6brich et al 1993). While the first one is a well-known standard procedure, the second is rather new and deserves a few words of explanation.…”

“…For particulate radiation, the yield of DSB s per unit dose has been found to increase with LET up to about 150 keV/gm in yeast cells (Frankenberg et al 1981;Akpa et al 1992;L6brich et al 1993) as well as in earlier studies with mammalian cells (Kampf 1982;Kampf and Eichhorn 1983). Such a LET-RBE (relative biological effectiveness) relationship is consistent with computational Monte-Carlo simulations Chatterjee and Holley 1991).…”

“…The initial yield of DSBs per unit dose has been found to depend on the quality of the radiation. For a particles, an increase in the yield of DSBs with linear energy transfer (LET) up to about 150 keV/pm has been found using some experimental systems (4)(5)(6), including mammalian cells in early studies (7,8). Such a behavior is also predicted by Monte Carlo simulations in computational studies (9,10).…”

“…Switching time was ramped from 30 s to 100 s over a 24 h running time. After electrophoresis, the gels were stained and destained following a procedure described by Lobrich et al (1993). The gel image was photographed under UV light with a polaroid camera.…”
