DNA metabarcoding Uncover the Diet of Subterranean Rodents in China

Abstract: Objective: In the past, the zokor, which lived in northern China, caused great harm to agriculture and forestry production due to its large and sophisticated diet. Since the rat lives underground for most of its life, researchers know little about its dietary habits. Further understanding of its diet in the field is of important meaning for developing green and sustainable control strategies for the rat. Methods:Longde County in Liupan Mountain area of Ningxia Hui Autonomous Region was selected as the inte… Show more

“…The RDP classifier bayesian algorithm version 2.11 ( ) was used on the QIIME (QIIME version 1.9.1) platform for classification analysis of representative sequences of OTUs from the sliva138/16S bacterial classification database with a default confidence threshold of 0.7. Core species analysis was used to indicate that the sample size was sufficient ( 27 ). The fecal alpha diversity among the two groups was evaluated via Sobs, Shannon and Simpson indices.…”

“…Generally, the lower parts of the forbs are enlarged and their nutrient content is more abundant. The distribution of the biomass of forbs determines the population density of plateau zokors in different regions to a certain extent (Zhang et al, 2010(Zhang et al, , 2022. Plateau zokors at different degradation levels and RG feed on the same species of plants but with different proportions.…”

“…During the last decade, DNA metabarcoding for dietary analysis has become a popular approach (Taberlet et al., 2018 ), mainly due to its cost efficiency and superior taxonomic resolution compared to morphological methods (da Silva et al., 2019 ; Soininen et al., 2009 ; Valentini et al., 2009 ). Thus, it has become a common method for resolving the diverse diets of small rodents (Aylward et al., 2022 ; Lopes et al., 2020 ; Sato et al., 2019 ; Zhang & Han, 2021 ). DNA metabarcoding also has the potential to provide information about the relative biomass proportions of ingested food via different approaches (Deagle et al., 2019 )—though neither is optimal in every study system (if in any).…”
