DNA Vaccine Combinations Expressing Either Tissue Plasminogen Activator Signal Sequence Fusion Proteins or Ubiquitin-Conjugated Antigens Induce Sustained Protective Immunity in a Mouse Model of Pulmonary Tuberculosis

Abstract: DNA vaccination has emerged as a powerful approach in the search for a more efficacious vaccine against tuberculosis. In this study, we evaluated the effectiveness of immunizing with combinations of 10 different tuberculosis DNA vaccines that expressed mycobacterial proteins fused at the N terminus to eukaryotic intracellular targeting sequences. In one vaccine combination, the genes were fused to the tissue plasminogen activator signal sequence (TPA), while in a second combination the same 10 genes were expre… Show more

“…1) suggests adding this sequence to all of our 4pox vaccine targets would be beneficial. This is further supported by numerous reports demonstrating the positive effects the tPA leader sequence can have on other antigens [43][44][45][46][47][48]. However, the ability of the tPA leader sequence to enhance neutralizing antibody production appears to be antigen specific.…”

“…To overcome this, we constructed a plasmid containing the L1R open reading frame (including the transmembrane region) fused to an up stream tissue plasminogen activator secretion signal sequence (TPA). This sequence has been used previously to target proteins through the endoplasmic reticulum and to the secretory pathway [16][17][18]. Because TPA-L1 retained its transmembrane domain, it was expected to localize to the plasma membrane of transfected cells.…”

“…The majority of TB antigens have been tested or screened in various prophylactic settings, whereas a relatively small number have been studied in therapeutic models. [4][5][6] Ag85A, 7 PstS3, 8 MPT64, 9 ACR, 10,11 Mtb39,12 Mtb72F (a fusion protein of Mtb32 and Mtb39) 13 and TB10.4 (ref. 14) were examined in this study.…”
