DOC-2/hDab2, a candidate tumor suppressor gene involved in the development of gestational trophoblastic diseases

Abstract: Gestational trophoblastic diseases comprise a spectrum of interrelated diseases including partial mole, complete mole and gestational choriocarcinoma. Using reverse transcriptase PCR (RT ± PCR) analysis, we identi®ed higher levels of DOC-2/hDab2 expression in the normal trophoblast cells in culture than in choriocarcinoma cell lines. Subsequent study using immunohistochemistry showed high levels of DOC-2/hDab2 protein expression in normal trophoblast tissues but signi®cantly lower levels of expression in gesta… Show more

“…Because this complex is important for the activation of K-ras, the dissociation of Sos/Grb2 may lead to decreased activation of K-ras. Transfection of a full-length DOC-2 cDNA construct into ovarian cancer and choriocarcinoma cell lines significantly reduces their growth rate, both in vitro and in vivo, suggesting a potential function of DOC-2 as a tumor suppressor (Fulop et al, 1998;Mok et al, 1998). However, the importance of DOC-2 in pancreatic cancer, which exhibits a high rate of K-ras Received February 14, 2001.…”

“…The mitogen-responsive phosphoprotein DOC2/DAB2, it has been suggested, is an essential component of the TGFbeta signalling pathway (Hocevar et al, 2001). Silencing of the gene has been described in ovarian, choriocarcinoma, pancreatic, prostate, and mammary carcinomas (Fulop et al, 1998;Huang et al, 2001;Mok et al, 1998;Tseng et al, 1999).…”

“…Similar to other human cancers, neoplastic transformation of trophoblast is likely to be a multistep process and involves multiple genetic alterations including activation of oncogenes and inactivation of tumor suppressor genes. 3,4 Potential biological markers related to malignant transformation of trophoblast include c-erbB-2, 5 cyclin E, 6 DOC-2/hDab2, 7 Ras GTPase activating protein, 8 telomerase activity, 9 apoptotic index, 10,11 and matrix metalloproteinases, 12 but no molecular parameter can currently replace serial serum human chorionic gonadotrophin (hCG) titers as the mainstay for predicting gestational trophoblastic neoplasia.…”
