Does autophagy have a license to kill mammalian cells?

Abstract: Macroautophagy is an evolutionarily conserved vacuolar, self-digesting mechanism for cellular components, which end up in the lysosomal compartment. In mammalian cells, macroautophagy is cytoprotective, and protects the cells against the accumulation of damaged organelles or protein aggregates, the loss of interaction with the extracellular matrix, and the toxicity of cancer therapies. During periods of nutrient starvation, stimulating macroautophagy provides the fuel required to maintain an active metabolism … Show more

“…In light of catabolism, these degenerative changes to connective tissues may be due to absorption of lipid content or other nutriment, which therefore is under study. Inanition is suspected, since chronic restrictions of other basic needs essential for survival result in the body's use of its own substrates to help meet the missing requirements (12,29,65).…”

“…However, it has only been shown in very few examples that autophagy genes are required for cellular death in animals. 4,40,41 In plants there is as yet also very little evidence for developmental PCD that depends on autophagous structures. 42,43 The evidence in plants rather suggests that knockout of ATG genes, thought to be associated with microand macroautophagy, does not inhibit but rather accelerates the visible PCD symptoms.…”

“…2,4 Both in animals [5][6][7] and yeast, 8 good ultrastructural evidence exists for the presence of autophagosomes. In root-tip meristem cells of various species, the formation of structures that are similar to autophagosomes/autolysosomes in animal cells has been documented in an excellent series of papers (reviewed in ref.…”

“…Although stimulation of autophagy might constitute a protective response, extensive and non regulated autophagy may lead to cell death which may be either independent or linked to apoptosis [11].…”
