Dorso-ventral axis perturbation in goldfish embryos caused by heat- and pressure-shock treatments for chromosome set manipulation

Abstract: Effects of the chromosome set manipulation on early embryonic development were examined ,in goldfish and crucian carp. Both pressure-and heat-shock treatments of fertilized eggs induced .developmental disorders such as thin blastodisc formation, delay of epiboly, and deficiency of dorso-anterior structures. The longer the treatments were applied to the eggs, the more ·frequently severe dorsal deficiencies were induced in the embryos. No and reduced signals of goosecoid mRNA, a dorsal mesodermal marker, were ob… Show more

“…In the goldfish and crucian carp ( Carassius auratus Linnaeus 1758), high pressure shock applied to the fertilized eggs results in the formation of the thin blastodiscs with poorly developed cytoplasm, delay of epiboly and suppression of the dorso ventral differentiation (Yamaha et al. ). Furthermore, high pressure that has been proved to damage DNA‐protein structure and affect gene expression (Fernandes et al.…”

“…However, high hydrostatic pressure shock, apart from destabilization of the microtubules can induce deformations of other cell organelles, disintegrate DNA-protein structure and change expression of the genes (Parkkiren et al 1993;Crenshaw et al 1996;Debec and Marcaillou 1997;Fernandes et al 2004). High pressure shock may disturb the dorso-ventral differentiation of the fish embryos when applied to the zygotes (Yamaha et al 2002). Moreover, we cannot exclude that the high hydrostatic pressure is damaging for the cytoplasmic mRNA and proteins indispensable for the proper embryonic development in the fish.…”

“…7). Abnormal medaka embryos produced by ICSI also resemble fish embryos deficient in the body axis, which were produced by disruption of microtubules by cold, heat, or pressure shocks [31], or by the drug nocodazole [32]. In zebrafish, a microtubule-dependent localization of the diffusible morphogen, Squint, to the blastodisc is required to establish the dorso-ventral axis in early cleavage stages [33,34].…”
