Draft Genome Sequence of Helicobacter suis Strain SNTW101, Isolated from a Japanese Patient with Nodular Gastritis

Abstract: We present here the draft whole-genome shotgun sequence of an uncultivated strain SNTW101 of Helicobacter suis, which has been maintained in the stomachs of mice. This strain was originally isolated from gastric biopsy specimens of a urea breath test-negative Japanese patient suffering from nodular gastritis.

“…The first one was made against the homogenate of cultured Hp and reported to react with Helicobacter species, including Hs and Hhss/Ha 13 . The second one was made against the synthetic peptide from the sequence of Hs‐specific outer membrane protein, VacA paralog (HsvA), which is well‐conserved among all the strains of Hs isolated from humans, pigs, and monkeys 14,20,21 . After washing with PBS three times for 5 minutes, we then reacted the specimens with a goat anti‐rabbit IgG conjugated with Alexa Fluor 488, (sb150077, abcam), washed them with PBS three times for 5 minutes, then counterstained with phalloidin‐iFluor reagent (ab176757, abcam) (Alexa Fluor 594) (both of which were diluted 1000 times with PBS) for 30 minutes, and washed with PBS three times for 5 minuutes.…”

“…We thus designed the present study to determine the more precise significance of NHPH infection in non‐Hp‐eradicated or post‐Hp‐eradicated human gastric diseases in Japan as revealed by polymerase chain reaction (PCR). In the NHPH‐positive cases, we used polyclonal antibodies against Helicobacters 13 and a Helicobacter suis (Hs) VacA paralog 14 to examine the NHPH immunohistochemically.…”

“…For immunohistochemistry of H. suis , rabbit polyclonal antibody against H. suis was made. Briefly, a rabbit was immunized with a peptide expected to be antigenic from the vacA paralog reported in a literature 19 . Rabbit IgG was purified from the serum with Protein G affinity column.…”

“…We have not successfully cultivated H. suis strains in vitro at this time. Therefore, the H. suis strains TKY (a highly pathogenic strain for mice) and SNTW101 used in this study were isolated from a cynomolgus monkey and a Japanese patient with nodular gastritis, respectively, and individually maintained in the stomachs of C57BL/6 mice (Charles River Japan, Yokohama, Japan) by repeated inoculations of gastric mucosal homogenates from infected to uninfected mice at intervals of around 6 months …”
