Drug delivery by erythrocytes: “Primum non nocere”

Abstract: Red blood cells (RBCs) are naturally capable of transporting diverse cargoes throughout the circulatory system, both loaded to their surface or within their inner volume. Starting largely from the 1970s, diverse approaches for encapsulation into, and surface coupling onto, RBCs have been investigated as potential drug delivery systems. In the last decade, these efforts have yielded diverse strategies to load drugs and nanocarriers to RBCs, and to optimize their pharmacokinetics, distribution, and effects in th… Show more

“…The emergence of stem-cell derived RBCs provides further opportunities for developing customized antibody delivery systems for immune therapy [13]. Currently, several drug-loading techniques such as encapsulation and surface conjugation have been used for the effective treatment of infections, cancer, and chronic autoimmune diseases [13]. More recently, there is progressive evolution in the use of inhalable drug delivery systems for lung cancer therapy.…”

“…The entrance of MPPs alters the membrane, making it susceptible to physiological and pathological changes that RBCs generally confront while circulation. Majority of MPPs are cationic in nature which may interact with glycocalyx of RBCs thereby, producing undesirable effects such as pro-inflammatory response to endothelium (Villa et al 2016).…”
