Drug Derived Fluorescent Probes for the Specific Visualization of Cannabinoid Type 2 Receptor - A Toolbox Approach

Abstract: Cannabinoid type 2 receptor (CB2R) is a fundamental part of the endocannabinoid signaling system (eCB system), and is known to play an important role in tissue injury, inflammation, cancer and pain. In stark contrast to its significance, the underlying signaling mechanisms and tissue expression profiles are poorly understood. Due to its low expression in healthy tissue and lack of reliable chemical tools, CB2R visualization in live cells remains uncharted. Here we report the development of a drug derived toolb… Show more

“…The workflow of this approach is shown in Figure 2. First, we chose a set of parameter values and random effects that reflect those from the real‐world experiment referred to above (Thais et al, 2019). Second, we propose several experimental designs to be tested, each with varying characteristics, including group sizes ( n ).…”

“…The Carreira group focussed on modifying pyrazine and pyridine scaffolds, in which no previous cannabinoid fluorescent probes have been published. [139] With parent compounds RO6839251 (K i (hCB 2 R) ¼ 0.2 nM) and RO6852763 (K i (hCB 2 R) ¼ 0.2 nM), displaying good affinity and selectivity to CB 2 R, fluorescent moieties NBD, AttoThio12, Silicon Rhodamine (SiR), and Cy5.5 were linked via variations on PEG or C-6 alkyl chains. NBD (59) and AttoThio12 (60) linked compounds displayed the highest affinity to CB 2 R but maintained moderate affinity to CB 1 R. In flow cytometry experiments, AttoThio12 and SiR (61) bound compounds labelled mouse and human CB 2 R, CHO cells overexpressing cannabinoid receptors, as well as WT CHO cells.…”
