Dual-colour CISH is a reliable alternative to FISH for assessment of topoisomerase 2-alpha amplification in breast carcinomas

Abstract: Anthracyclines are among the most powerful antineoplastic drugs available for breast cancer treatment. Although HER2 amplification has been postulated to predict anthracycline benefit, numerous reports have demonstrated that HER2/TOP2A co-amplification is the clinically useful predictive marker of response to anthracyclines. The standard technique to evaluate gene status for target therapy selection is fluorescence in situ hybridization (FISH), but this technique has some disadvantages. Dual-colour chromogenic… Show more

“…The identified studies were classified either as early or mainstream HTA, but none as very early HTA. Almost all early HTA articles reported on the comparative effectiveness of testing techniques [41][42][43][44][45]. Only one article presented an early stage cost-effectiveness analysis [46].…”

“…The construction of TMAs involves the repeated transfer of small tissue cores, typically ranging from 0.6 to 2.0 mm in diameter, from para n-embedded tissue blocks into a single empty para n block [14,16]. TMAs are an ideal common tool for highthroughput pro ling to study tissue morphology, protein or gene expression following different stains, immunohistochemistry or uorescence in situ hybridization [16,17]. Because of their inherent characteristics, large numbers of samples can be tested and analyzed in a single running.…”
