Dynamic MR imaging and tumor angiogenesis in DMBA-induced rat breast cancer: Three dimensional (3D) reconstructed image analysis of tumor microvessels by confocal laser scanning microscopy

Abstract: We investigated the correlation between dynamic magnetic resonance imaging (MRI) findings in breast cancer and tumor angiogenesis in a dimethylbenz(a)anthracene(DMBA)-induced rat breast cancer model. In this study, we clearly demonstrated the three-dimensional (3D) architecture of tumor microvessel networks(MN)by confocal laser scanning microscopy (CLSM)and also investigated the hyperpermeability of tumor microvessels. Dynamic MRI findings were closely related to tumor angiogenesis. Three-dimensional reconstru… Show more

“…For instance, Itoh et al (46) were able to obtain three-dimensional reconstructed images of cells in the pituitary gland and their microvascular environment using confocal laser microscopy. Yasumura et al (105) were able to determine by confocal laser microscopy microvessel densities and permeabilities in three-dimensional reconstructed images of rat mammary tumor. In the kidney, the capillary network of the glomerulus (50) and its circulation (70) have been studied.…”

“…( 28,29 ) It should be emphasized that the use of confocal laser scanning microscopy (CLSM) together with MRI may have great potential to evaluate antivascular agents in tumor regions in more detail, because the tumor vessel network and hemodynamics can be examined directly. ( 26 )…”

“…The antivascular effect of TZT‐1027 was evaluated using MRI technique using a 1.5 Telsa system (Signa echo speed, GE Medical Systems, WI, USA), as reported previously. ( 26 ) After anesthesia with pentobarbital was initiated, the tumor‐bearing rats were placed on a phased array extremity coil designed for knee examination. A 27 G butterfly needle was inserted into the tail vein of each rat for remote injection of the contrast medium.…”
