E. coli Secretome Metabolically Modulates MDA-MB-231 Breast Cancer Cells’ Energy Metabolism

Abstract: Breast cancer (BC) is commonly diagnosed in women. BC cells are associated with altered metabolism, which is essential to support their energetic requirements, cellular proliferation, and continuous survival. The altered metabolism of BC cells is a result of the genetic abnormalities of BC cells. Risk factors can also enhance it, including age, lifestyle, hormone disturbances, etc. Other unknown BC-promoting risk factors are under scientific investigation. One of these investigated factors is the microbiome. H… Show more

“…Quality control samples (QCs) were performed gently by collecting a single 3.2 mm punch from each study sample and pooling them for extraction. After that, they were introduced to the instrument randomly to validate the system’s stability [ 58 ]. After that, they were analyzed following the routine protocol.…”

“…To investigate the metabolic alterations occurring in our cell model, we followed a previously reported protocol to extract intra- and extracellular metabolites [ 10 ]. Briefly, in intracellular metabolism, medium was removed, and cells were washed with cold PBS followed by quenching in liquid nitrogen.…”

“…The samples were reconstituted in 50% mobile phase A:B (A: 0.1% formic acid in dH 2 O, B: 0.1% formic acid in 50% MeOH and ACN) for untargeted metabolomics analyses using LCMS as previously reported [ 10 ]. Metabolites were acquired by a Waters Acquity ultra pressure liquid chromatography (UPLC) system coupled with a Xevo G2-S QTOF mass spectrometer equipped with an electrospray ionization source (ESI) on positive and negative (ESI+, ESI−).…”

“…Metabolomics profiling was performed using untargeted metabolomics analyses by LC-HRMS, as previously reported [22]. Metabolites were acquired by a Waters ACQUITY ultrahigh-pressure liquid chromatography (UPLC) system coupled with a Xevo G2-S QTOF mass spectrometer equipped with an electrospray ionization source (ESI) in positive and negative modes (ESI+, ESI−).…”
