Early Growth Response-1 Plays a Non-redundant Role in the Differentiation of B Cells into Plasma Cells

Abstract: Early growth response (Egr)-1 is a Cys2-His2-type zincfinger transcription factor. It has been shown to induce survival and proliferation of immature and mature B cells, respectively, but its role in the differentiation of B cells into plasma cells remains unclear. To examine the effects of Egr-1 deficiency on the activation of B cells, naive B cells from Egr1-/- mice and their wild-type (WT) littermates were activated to proliferate and differentiate, and then assayed by FACS. Proportions of cells undergoing … Show more

“…The EGR1 binding box was previously demonstrated necessary for BLIMP1 expression [46]. Recently in an in vivo study, egr1 −/− mice have impaired ASC differentiation yet with normal B cell development and intact cell proliferation and apoptosis [57]. This feature is highly matching what we have observed in iguratimod-treated human B cells.…”

“…Recently, B cells lacking Egr1 were shown to undergo reduced differentiation to CD138 + plasmablasts when cultured with LPS (117). Furthermore, Egr1 knockout mice secrete less antigen-specific antibody in response to immunization with a T-dependent antigen (117). Since Egr1 is overexpressed in Ets1 −/− B cells, reducing the levels of this gene might be expected to reverse the phenotype of Ets1 −/− B cells and reduce ASC numbers.…”
