EBF1 drives hallmark B cell gene expression by enabling the interaction of PAX5 with the MLL H3K4 methyltransferase complex

Abstract: PAX5 and EBF1 work synergistically to regulate genes that are involved in B lymphocyte differentiation. We used the KIS-1 diffuse large B cell lymphoma cell line, which is reported to have elevated levels of PAX5 expression, to investigate the mechanism of EBF1- and PAX5-regulated gene expression. We demonstrate the lack of expression of hallmark B cell genes, including CD19, CD79b, and EBF1, in the KIS-1 cell line. Upon restoration of EBF1 expression we observed activation of CD19, CD79b and other genes with … Show more

“…7c). Twenty-four percent of total B cellspecific TFs are shared between the two organisms including Tcf4 [38][39][40][41], Pax5 [78][79][80], Bhlhe41 [81][82][83], and several Irf family TFs (Irf4, Irf5, Irf8) [24,84,85]. This relatively large overlapping percentage is consistent with the clustering of B cells across species shown in Fig.…”

“…Although MEF2D mutation results in maturation arrest at the pre-B stage, its distinct immunophenotype is recognized to overlap with both early and late B progenitor stages 20 . The similarity of cell signals in PAX5 and KMT2A mutant B-ALL may represent the intimate relationship of KMT2A and PAX5 in regulating B lymphopoiesis 21 .…”

“…Presence of PAX5 on chromatin correlated with increased active chromatin marks in PAX5induced genes, whereas an inverse pattern of histone modifications was observed in PAX5-repressed genes [281]. As demonstrated later, another B-cell specific transcription factor, EBF1, is required for the interaction of PAX5 with the MLL H3K4 methyltransferase complex and subsequent epigenetic modifications [282]. EBF1 and PAX5 have opposing roles in normal and malignant B cells with regards to the regulation of the MYC oncogene.…”
