Ectonucleotidase activities in Sertoli cells from immature rats

Abstract: Sertoli cells have been shown to be targets for extracellular purines such as ATP and adenosine. These purines evoke responses in Sertoli cells through two subtypes of purinoreceptors, P 2Y2 and P A1 . The signals to purinoreceptors are usually terminated by the action of ectonucleotidases. To demonstrate these enzymatic activities, we cultured rat Sertoli cells for four days and then used them for different assays. ATP, ADP and AMP hydrolysis was estimated by measuring the Pi released using a colorimetric met… Show more

“…Sertoli cells were isolated from the testis of 18-day-old Wistar rats by sequential enzymatic digestion, as described by Casali et al (2001). The testes were sequentially digested with 0.25% trypsin and DNase (10 μg/ml) for 30 min at 37°C to remove the interstitial tissue.…”

“…The Sertoli cell monolayers were incubated as previously described by Casali et al (2001). Briefly, the Sertoli cell monolayers were washed 3 times with the reaction medium containing 135 mM NaCl, 5 mM KCl, 10 mM glucose and 10 mM HEPES, 2 mM CaCl 2 pH 7.4.…”
