This study analyzes the action of sulfated polysaccharides, fucans, from algae Lobophora variegata on zymosan-induced arthritis in rats. Groups of fucans, obtained after acetone fractionation (0.3-2.0 volumes), were denominated F0.3, F0.5, F0.8, F1, F1.5, and F2. The results that F1 contained a high yield in relation to other fractionated fucans. Chemical and structure analysis of F1 was performed by nuclear magnetic resonance (NMR) and infrared (IR) spectroscopies. The in vitro antioxidant activities of the fraction F1 were also observed. Thus, 2 mg/mL of F1 inhibited the phosphomolybdate in the total antioxidant activity assay. The EC(50) values were 0.3 mg/mL and 0.12 mg/mL for superoxide and hydroxyl radicals, respectively. Fucan F1 (25, 50, and 75 mg/kg by body weight), diclofenac sodium (10 mg/kg), and L-NAME (25 mg/kg) were administered intraperitoneally (i.p.) in rats, according to body weight of different groups of animals (n=6). After 6 h, analyses of cell influx and nitrite levels were conducted. Then after 96 h, analysis of edema and concentration of serum TNF-α was carried out along with histopathological analysis. F1 at 25, 50, and 75 mg/kg i.p. by body weight reduced cell influx in 52.1-96.7% and nitric oxide level in 27.2-39% compared with the control group. The reduction of edema and serum TNF-α was observed at 50 mg/kg i.p. (p<0.001). These results suggest that this heterofucan from the brown algae L. variegata has potential anti-inflammatory activity in acute zymosan-induced arthritis in rats and that antioxidant activity promotes modulation in the cellular redox state.

Studies have demonstrated the beneficial effects of fangotherapy on relieve of pain improving function of rheumatic patients. Herein, we investigated the effect of Brazilian black mud in protect articular damage in chronic arthritis induced in rats. Mud was daily applied (40 degrees C/30 min) during the course of arthritis and was compared with warm water and no treated groups. At 21th day after arthritis induction synovial fluid and membrane were analyzed regarding cellular influx, hyperplasia and vascular proliferation. Cartilage structure, cell count, proteoglycan and collagen amount were also analyzed by three pathologists blinded to the treatment. Mud treatment diminished leukocyte migration into the synovial membrane and articular cavity when compared with both control groups. Regarding cartilage, an increase in collagen, number of chondrocytes and more conserved tissue structure was observed in mud-treated animals. These results demonstrate a protective effect of Brazilian mud on this model of arthritis, suggesting that this therapy may be useful as a complementary approach to treat articular diseases.

The aim of this study was to evaluate the effect of low-level laser therapy on acute zymosan-induced arthritis, with respect to the laser action on inflammatory cells influx, release of pro-inflammatory mediators, metalloproteinases activity into the joint cavity and the cartilage repair process. Arthritis was induced in male Wistar rats (250-280 g) by intra-articular injection of zymosan (1 mg dissolved in 50 μl of a sterile saline solution) into one rear knee joint. Animals were irradiated immediately, 1 and 2 h after zymosan administration with a semiconductor laser InGaAIP (660 nm, 10 mW, 2.5 J/cm(2), 10 s). In the positive control group, animals were injected with the anti-inflammatory drug dexamethasone 1 h prior to the zymosan administration. Treatment with laser significantly inhibited leukocytes influx, the release of IL-1 and IL-6 and also the activity of metalloproteinase-2 and 9, into the joint cavity. In conclusion, laser therapy was effective in reducing inflammation to sites of injury and inhibit activation of proteases (gelatinase) suggesting less degradation of collagen tissue in experimental model of acute arthritis.
