Effect of Dietary Silk Peptide on Obesity, Hyperglycemia, and Skeletal Muscle Regeneration in High-Fat Diet-Fed Mice

Abstract: Obesity is associated with excess body fat accumulation that can cause hyperglycemia and reduce skeletal muscle function and strength, which characterize the development of sarcopenic obesity. In this study, we aimed to determine the mechanism whereby acid-hydrolyzed silk peptide (SP) prevents high-fat diet (HFD)-induced obesity and whether it regulates glucose uptake and muscle differentiation using in vivo and in vitro approaches. Our findings demonstrate that SP inhibits body mass gain and the expression of… Show more

“…Targeting energy expenditure represents an interesting concept as a means of combating obesity. The discovery of adipose browning and its associated lipid metabolism has generated interest in the search for natural dietary compounds that could be a therapeutic strategy for prevent obesity and its associated metabolic syndrome [33,34]. In the present study, we evaluated two mechanisms whereby ESE might prevent obesity, suppress fat accumulation, and increase energy expenditure-the upregulation of lipolysis and the induction of browning, using HFD-induced obese mice and 3T3-L1 cells, respectively.…”

“…Obesity occurs due to an imbalance between energy intake and energy expenditure [37]. WAT accumulates excessive energy in the form of TGs, which is directly associated with obesity.…”

“…Thus, promoting a BAT-like phenotype in WAT, known as WAT browning, could increase energy dissipation. A variety of dietary molecules, such as berberine, ginsenoside Rg1 and silk peptide, are reported to modulate adipocyte expression of thermogenic transcription factors [37,38]. In the present study, we aimed to investigate the anti-obesity effects of GN in the context of AMPK-mediated lipid metabolism and adipocyte browning.…”

“…The free amino acid composition of SP was analyzed using an HPLC system comprising a pump (Waters 2695, Waters, Milford, MA, USA), an AccQ-Tag amino acid analysis column (Silica C18, 3.9 mm × 150 mm), and a Waters 2475 Multiλ Fluorescence detector (24,25). The HPLC chromatogram is shown in Supplementary Data 3 in Supplementary Material.…”
