Electron paramagnetic resonance (EPR) was used as a method for recording the content of the nitric oxide (NO) in hippocampal tissues of intact rats and rats after modelling of ischaemic and haemorrhagic stroke. Based on direct measurements of NO by EPR spectroscopy, it was shown that, within 5 hours after the onset of symptoms of ischaemic and haemorrhagic stroke, the formation of NO in the hippocampus was reduced by a factor of 2-3 and this reduction was maintained for a period of between 24 and 72 hours. The results show that a systemic character of a decrease in the intensity of NO production during the modelling of ischaemic events in the brain reflects the effects of central dysregulation of the functions at the level of the whole organism such that it is appropriate to consider implementing the correction of the vital systems of the body in a stroke. It has indicated that non-selective NO-synthase blocker L-NAME reduced the low level of NO production by a factor of 3 by its administration within 72 hours after post-ischaemic and haemorrhagic stroke. It was discovered however that L-NAME returns the level of NO production to baseline (control) by its administration within 5 hours after ischaemia.

На експериментальній моделі індукованого гліцерином рабдоміолізу в щурів досліджено утворення нового пулу негемового феруму в плазмі крові та з'ясована його причетність до інтенсифікації утворення NO. Вже на першу добу після внутріш-ньом'язового введення тваринам 50% гліцерину в дозі 10 мл/кг реєструється 10-кратне збільшення вмісту вільного гему у плазмі крові та 6-кратне підвищення гемоксигеназної активності у печінці. За допомогою методу ЕПР доведено факт утворення у плазмі крові дослідних тварин пулу лабільного тривалентного феруму, який був повністю відсутнім у зразках інтактної групи. У період з першої по шосту добу досліду вміст цієї форми феруму становив 2 мг/л, що суттєво перевищувало вміст заліза у трансферині. На фоні формування пулу лабільного феруму крові спостерігалося підвищення показників нітрозативного стресу. Так, на четвертий день спостерігалося збільшення вмісту загальної NO синтази щодо контролю у печінці та S-нітрозотіолів і нітрозопроліну в плазмі крові удвічі. Для перевірки гі-потези про причетність лабільного феруму до розвитку нітрозативного стресу був використаний унітіол як акцептор феруму. Застосування унітіолу привело до змен-шення концентрації лабільного феруму в плазмі крові утричі на четверту добу роз-витку патології порівняно з тваринами з експериментальним рабдоміолізом. Ви-явлено позитивну кореляцію між вмістом лабільного феруму в крові та показниками нітрозативного стресу: вміст загальної NO синтази, нітрозопроліну та S-нітрозо тіо-лів зменшився у 1,5 разу на фоні падіння концентрації лабільного феруму в крові. За умови розвитку рабдоміолізу, стимульованого гліцерином, у плазмі крові щурів відбувається утворення нового пулу негемового тривалентного феруму, яке не зв'я-зане з трансферином і тому здатне до хімічної взаємодії. Таким чином, лабільний ферум є одним із ініціаторів нітрозативного стресу при рабдоміолізі.Ключові слова: ферум, трансферин, рабдоміоліз, унітіол, NO.
