Effect of Donor Cell Type on Nuclear Remodelling in Rabbit Somatic Cell Nuclear Transfer Embryos

Abstract: Cloned rabbits have been produced for many years by somatic cell nuclear transfer (SCNT). The efficiency of cloning by SCNT, however, has remained extremely low. Most cloned embryos degenerate in utero, and the few that develop to term show a high incidence of post-natal death and abnormalities. The cell type used for donor nuclei is an important factor in nuclear transfer (NT). As reported previously, NT embryos reconstructed with fresh cumulus cells (CC-embryos) have better developmental potential than those… Show more

“…Like pigs, rabbits also have a more diverse genetic background than rodents, a situation that is closer to that in humans. Notably, the first transgenic rabbits were generated over 3 decades ago (37, 38), but the lack of bona fide rabbit ESCs for more complex genetic engineering and the inefficiency of rabbit somatic cell nuclear transfer (39) hampered the development of the field until the arrival of highly efficient designer nuclease technologies (20, 4042). …”

“…In buffalos, cumulus cells are a more efficient nuclear donor for SCNT than fibroblasts [30]. In rabbits, embryos reconstructed with fresh cumulus cells have a more efficient developmental potential than those reconstructed with fetal fibroblasts both in vivo and in vitro [31]. However, comparisons show that adult cells of any type are inferior to fetal fibroblasts in terms of reconstructed embryo development.…”

“…Nevertheless, neither the donor cell type nor the gender significantly affects the overall efficiency of the in vitro production of SCNT sheep embryo [33]. In rabbit, embryos reconstructed with fresh cumulus cells have a more efficient developmental potential than those reconstructed with fetal fibroblasts in vivo and in vitro [34]. In pig, the type of donor somatic cell is important for the development of cloned embryos; fetal fibroblasts are the most effective among adult and fetal fibroblasts, cumulus and oviduct cells [22].…”
