Effect of ectopic high expression of transcription factor OCT4 on the “stemness” characteristics of human bone marrow-derived mesenchymal stromal cells

Abstract: Objective To investigate the effect of ectopic high expression of OCT4 on the stemness characteristics of bone marrow-derived mesenchymal stromal cells (BM-MSCs). Methods BM-MSCs were collected from three de novo acute lymphoblastic leukemia (ALL) and three aplastic anemia patients (AA), which were cultivated by the whole bone marrow adherent method. Surface markers of BM-MSCs were analyzed by flow cytometry (FCM); meanwhile, growth characteristics were observed with a … Show more

“…1b ). However, the expression of neural ectodermal and neural plate border markers, SIX1, TUBB3 and ZIC1 25 27 , was upregulated after induction. In addition, the expression of the mesodermal markers KDR, TBX6, and PAX1 28 – 30 and the endodermal markers GATA4, GATA6, and SOX7 31 , 32 were not significantly induced after treatment with CSSEDF medium (Supplementary Fig.…”

“…Overexpression of Oct4 improves proliferation capacity of cells and can upregulate expression of Nanog and Sox2 and primarily maintain the morphology of MSCs. Thus, possible, that similar methods can be used to obtain valuable MSCs for stem cells therapy [78].…”
