Effect of Phosphatidic Acid on Human Breast Cancer Cells Exposed to Doxorubicin

Abstract: We previously demonstrated that phosphatidic acid (PA) induces chemotactic migration of highly metastatic breast cancer cells MDA-MB-231. The widely used anticancer drug doxorubicin was reported to induce apoptosis of cancer cells. Growth factors such as epidermal growth factor (EGF) and bioactive lipids such as lysophosphatidic acid (LPA) and sphingosine 1-phosphate (SPP) have been shown to enhance viability and to protect cancer cells against apoptosis. In this study, we investigated the effect of PA on MDA-… Show more

“…After incubation, the cells were harvested by trypsinisation, washed with Dulbecco's phosphate-buffered saline containing 2% FBS, and resuspended in propidium iodine (50 mg ml À1 ). Cell cycle analysis was performed on a FACStar PLUS flow cytometer (BectonDickinson, San Jose, CA, USA), as previously described (Sliva et al, 2001). Data are the mean±s.d.…”

“…Fluorescence and phase contrast microscopy x 400. inhibition of PP1 [13] or that PA determines a range of effects associated with proliferation and cell survival (reviewed in [1]). Other studies give evidence of opposite effects, such as potentiation by PA of doxorubicin-induced apoptosis in breast cancer cells [14]. Dual effects of phosphatidic acid have also been described on smooth muscle contractility [15].…”

“…After treatment with increasing concentrations of H 2 O 2 , the percent of necrotic cells is dramatically increasing, but the percent of apoptotic cells remains low (12)(13)(14)(15)(16)(17), indicating that apoptosis is not the main mechanism of cell death [9]. Exogenous H 2 O 2 may produce necrosis by lipid peroxidation [10] and NF-B activation [11,12].…”
