This study was an examination of the effect of interleukin-1 alpha (IL-1 alpha) on prostaglandin (PG) production and cyclooxygenase (COX) activity in endometrial stromal cells isolated from uteri of ovariectomized rats sensitized for the decidual cell reaction. Treatment with IL-1 alpha (20 ng/ml) for 24 h caused approximately 13-, 2-, and 2-fold increases in PGE2, PGF2 alpha, and 13, 14-dihydro-15-keto-PGF2 alpha (PGFM) accumulation, respectively, in the medium. The IL-1 alpha-induced increase in PG accumulation was accompanied by a significant (6-fold) increase in COX activity in the cells. IL-1 alpha-induced increases in PGE2 accumulation and COX activity were greatly reduced or prevented by cycloheximide (CHX), actinomycin D, and dexamethasone (DEX). In addition to IL-1 alpha, epidermal growth factor (EGF, 40 ng/ml) also caused a significant increase in PGE2 accumulation in the medium and COX activity in the cells. The combined effects of IL-1 alpha and EGF on PGE2 accumulation and COX activity were additive. The results of this study provide evidence that IL-1 alpha stimulates PG production in endometrial stromal cells from uteri sensitized for the decidual cell reaction through a mechanism that involves an increase in COX activity.

This study addresses the regulation of the interleukin 1 (IL1) system in the murine uterine luminal epithelium (LE) and stroma by leukemia inhibitory factor (LIF). Using RT-PCR we compared expression of Il1a, Il1b, Il1rn, Il1r1, and Il1r2 during the pre- and peri-implantation periods of pregnancy in wild-type (WT) and LIF-null LE and stroma. In WT LE, Il1a transcripts were down-regulated on Day 4 of pregnancy (D4), with renewed expression by the evening of D4 (D4 pm). In Lif(-/-) LE there was a gradual decrease in expression on D2, and expression became undetectable by D6. Il1b and Il1r1 expression were similar in WT and null mice, but Il1rn expression was almost completely lost during the peri-implantation period in Lif(-/-) LE. In the stroma, Il1a was sharply down-regulated on D4 and reappeared on D4 pm but was only expressed from D3 to D5 in the null mice. Stromal Il1r1 and Il1r2 were also misregulated. Il1rn showed constitutive expression in null stroma in contrast to the loss of expression on D4 in the WT mouse. In Lif-deficient mice, immunostaining indicated a reduction of endometrial IL1A at the time of implantation and of IL1B in stroma. LE-stromal coculture revealed that LIF stimulated the apical secretion of both IL1A and PTGES2 by LE cells without affecting basal secretion of IL1A and with only a small effect on basal PTGES2 secretion. We conclude that Il1a and Il1rn in LE and Il1a, Il1rn, and Il1r1 in stroma are regulated by LIF, which stimulates apical secretion of IL1A by LE.

Prostaglandins (PGs) are critical regulators of a number of reproductive processes. To date, the presence and regulation of PGS in the rat endometrium have not yet been described. The objective of the present study was to investigate the expression of PGD synthase (PGDS) and prostacyclin synthase (PGIS) in the endometrium. Endometrial proteins and tissues were collected from cyclic non-pregnant, pregnant, and steroid-induced pseudopregnant rats. PGIS and PGDS were detected in the endometrium of cyclic, pregnant, and pseudopregnant rats but were not influenced by the estrous cycle. During early pregnancy, PGIS was significantly higher at day 5 and was gradually decreased from day 5 . 5 to 6 . 5. Later during pregnancy, PGIS was maximal on day 12 and gradually decreased to the end of pregnancy. PGDS expression was high during early and was maximal at the end of pregnancy. During pseudopregnancy, PGDS and PGIS were increased in a time-dependent manner and were maximal at day 5. Immunohistochemical analysis revealed that PGDS and PGIS were found in luminal as well as glandular epithelial cells and in stroma during late pregnancy. We also found a significant increase of PGD 2 serum metabolite at days 21 and 22 of pregnancy. During steroidinduced pseudopregnancy, PGI 2 serum metabolite was increased in a time-dependent manner and was maximal at day 7. These results suggest that PGDS and PGIS are present and could be regulated by steroids in the rat uterus during pregnancy, and that the endometrium could be a significant source of PGD 2 and PGI 2 at specific times during pregnancy.
