Astaxanthin is a high value keto‐carotenoid pigment renowned for its commercial application in various industries comprising aquaculture, food, cosmetic, nutraceutical and pharmaceutical. Among the verified bio‐resources of astaxanthin are red yeast Phaffia rhodozyma and green alga Haematococcus pluvialis. The supreme antioxidant property of astaxanthin reveals its tremendous potential to offer manifold health benefits among aquatic animals which is a key driving factor triggering the upsurge in global demand for the pigment. Numerous scientific researches devoted over a number of years have persistently demonstrated the instrumental role of astaxanthin in targeting several animal health conditions. This review article evaluates the current best available evidence to judge the beneficial usage of astaxanthin in aquaculture industry. Most apparent is the profound effect on pigmentation, where astaxanthin is frequently utilized as an additive in formulated diets to boost and improve the coloration of many aquatic animal species, and subsequently product quality and price. Moreover, the wide range of other physiological benefits that this biological pigment confers to these animals is also presented which include various improvements in survival, growth performance, reproductive capacity, stress tolerance, disease resistance and immune‐related gene expression.

Following a request from the European Commission, the FEEDAP Panel was asked to deliver a scientific opinion on the safety and efficacy of astaxanthin (ATX)‐rich Phaffia rhodozyma. The additive, belonging to the category ‘sensory additives’ and the functional group ‘substances which, when fed to animals, add colours to food of animal origin’ is intended to be used in feed for salmon and trout from an age of six months onwards up to a maximum content of 100 mg ATX/kg complete feed. The product is produced by the telemorph of Phaffia rhodozyma, Xanthophyllomyces dendrorhous, and it is declared to contain 995 g dried inactivated biomass and 5 g ascorbic acid per kg additive. The main active principle of the additive is ATX; however, the FEEDAP Panel noted that some other carotenoids are also present in lower quantities. The minimum ATX concentration is specified to be 5,000 mg per kg additive. The yeast Xanthophyllomyces dendrorhous is considered by EFSA to be suitable for the qualified presumption of safety (QPS) approach to safety assessment; therefore, the use of the production strain in the production of the additive would not raise any safety concern for the target species, the consumers of products from animals fed the additive and the environment. In the absence of a tolerance study with the additive, the FEEDAP Panel cannot conclude on the safety for the target species. In the absence of residue and toxicity data of ATX, no final conclusions on the safety for the consumer can be drawn. The FEEDAP Panel concluded that the additive is irritant to skin and eyes, and a skin and respiratory sensitiser, although exposure by inhalation is likely low. The FEEDAP Panel considers that ATX from the biomass does not pose a significant additional risk to the environment compared with other natural sources of ATX. In absence of adequate evidence, no conclusion can be made on the efficacy of the additive.

This study aimed to evaluate the effect of astaxanthin on the hematological, biochemical and somatic response of Nile tilapia (Oreochromis niloticus) orally administered in the feed for a period of 60 days. For the study, 105 tilapia (n=35) from the same spawn were used, constituting the following treatments: Control = animals (not treated with astaxanthin); T100 and T200 = fish treated with 100 and 200 mg of astaxanthin/kg of feed, respectively. There were no hematological alterations in red blood cells counts, hematocrit, hemoglobin, MCV and MCHC. In addition, astaxanthin-fed tilapia presented a better thrombocyte and leukocyte responses with a marked decrease in the number of thrombocytes, lymphocytes and neutrophils. Among treatments, there were no changes in serum levels of total protein, albumin, globulins, aspartate aminotransferase and alkaline phosphate. Treatment with astaxanthin resulted in a decrease in triglyceride and glucose levels, as well as increase in hepatosomatic indices. The results of hematological and biochemical analyzes of tilapias demonstrated the clinical safety of this carotenoid, not causing harmful effects to the health of fish. Therefore, the antioxidant activity of this compound in tilapias resulted in an improvement in the leukocyte profile and contributed to hypolipidemic effects at the dose of 200mg of astaxanthin/kg of feed.
