Effect of the Antibiotic Neomycin on the Toxicity of the Glycoside Vicine in Rats

Abstract: Vicine is hydrolyzed by microflora to highly reactive free radical generating compound divicine which causes mortality and other adverse effects. This study in the rats established the effect of a broad spectrum and poorly absorbed antibiotic, neomycin sulfate on the toxicity of vicine. The results showed extremely decrease in mortality rate in the group pretreated with neomycin. Hemoglobin (Hb) concentration, hematocrit (Hct) value, and red blood cells (RBCs) count were significantly decreased after injection… Show more

“…The decline in WBCs was also observed by many researchers (Arbid and Marquardt,1986; Yanni and Marquardt,1985; Arbid et al , 2013) after the injection of vicine and convivine in male albino rats. On the other side, the oral administration with pickled olives+ V. faba in favism animals' models, pickled olives + vicine in vicine-treated animals and pickled olives + convicine in convicine-treated animals pushed WBCs to the control value.…”

“…The cleavage amplified polymorphism markers methods are used to produce faba beans cultivars with low vicine and convicine content and enhanced nutritional use of faba beans [7]. Antibiotic neomycin counteracts the acts of favism inducing factor ''vicine'' in rats [8]. Furthermore, favism induced oxidative stress and genotoxicity which abolished by anise oil treatment [9].…”

“…(2) Paraffin oil group: injected IP with 0.5 ml paraffin oil/rat and this oil was used a vehicle for vitamin E because this oil does not contain vitamin E or any antioxidant in contrast to many other oils such as olive oil, soybean oil, sunflower oil and corn oil which contain vitamin E. (3) Vicine and convicine (VC)-treated group: injected IP with 400 mg/kg V (Arbid et al, 2013) dissolved in 0.5 ml of distilled water and after 1 h the same rats injected IP with 150 mg/kg C (Arbid et al, 1993) dissolved in 0.5 ml of distilled water. (4) Vitamin E (100 mg/kg) þ VC-treated group: injected IP vitamin E (100 mg/kg) dissolved in 0.5-ml paraffin oil, 30 minutes before the injection IP of both V and C, daily once a day into VC-treated group.…”

“…So, an acute hemolysis caused by Vicia faba ingestion in G6PD deficient individuals (favism) is the best studied natural model of oxidative damage (Arese et al, 2012). The antibiotic neomycin and natural products such as anise oil, a-tocopherol, b -carotene and pickled olives were used to overcome the V and C toxicity (Arbid et al, 2013;Koriem et al, 2016Koriem et al, , 2017Koriem et al, , 2018Koriem and Arbid, 2020). Vicia faba contains V at 10-15 mg/g of dry weight and C at 3-5 mg/g of dry weight (Kowalczyk et al, 2020).…”
