Effect of Titanium Dioxide Nanoparticles on the Stereological Parameters of the Dentate Gyrus and the Morphology of Granular Hippocampal Neurons in Mice

Abstract: This study aims to investigate the Effects of Titanium dioxide nanoparticles (TiO 2 NPs) on the stereological parameters in the dentate gyrus and the morphology of granular hippocampal neurons in adult mice. Adult male mice (n=20, weight average: 45 g) were randomly divided into four groups including: group receiving saline (controls), low-dose (LD) 2.5 mg/kg TiO TiO 2 NPs, medium-dose (MD) 5 mg/kg TiO 2 NPs and high-dose (HD) 10 mg/kg TiO 2 NPs, daily using gavage for 35 days. To estimate the volume of the hi… Show more

“…In mice, the Panel noted that the reduced volume of the polymorph layer of the hippocampal dentate gyrus and reduced density and number of dentate gyrus granular neurons reported by Rahnama et al. (2020) with TiO 2 NPs (21 nm), is consistent with the behavioural effects reported by Zhang et al. (2020) also with TiO 2 NPs (21 nm), i.e.…”

“…The most sensitive endpoint observed in adult mice was reduced volume of the hippocampus, and the polymorph layer of the dentate gyrus, and reduced density and total number of dentate gyrus granular cells at all doses tested (in males dosed for 35 days) (Rahnama et al., 2020; scoring 4 for NSC). The mice in this study (n = 20/group) were dosed by gavage with TiO 2 NPs (21 nm) at 0, 2.5, 5 or 10 mg/kg bw per day.…”

In the hippocampus, different forms of TiO 2 -NPs were shown to exert some adverse effects such as oxidative stress, mitochondrial dysfunction, gliosis, and to provoke neuronal lesions [1,[28][29][30]. Exploiting reconstructed neuronal networks, TiO 2 -NPs were also reported to severely impair the electrical activity of neurons [31].…”
