Effectiveness of gene expression profiling for response prediction of rectal cancer to preoperative radiotherapy

Abstract: We identified radiation-related genes in colorectal cancer and demonstrated that PTMA may play an important role in radiosensitivity. Our findings suggest that PTMA may be a novel marker for predicting the effectiveness of radiotherapy in clinical cases.

“…Therefore, accurate selection of patients who are suitable candidates for neoadjuvant therapy will significantly improve the outcomes. Many molecular markers, including P53, p21, BCL2, BAX, EGFR, COX2, PTMA and ELF5a1, were identified before as predictors of response to this modality of treatment; however, their clinical application is still under evaluation [67,528]. Although no pre-treatment biopsies were used, the list of genes identified to be dysregulated in response to neoadjuvant therapy in this study is consistent with previous reports.…”

“…Moreover, stromal CXCL12 and CXCR4 expression was found to be associated with recurrence and poor survival after neoadjuvant therapy in the 53 patients analysed using RQ-PCR and immunohistochemistry [528]. However; comparison of expression levels of these genes in pre-and post-treatment biopsies is required to further validate their use as predictors of response.…”

“…An association with cancer has been reported for prothymosin-a (PTMA) that is involved in chromatin de-condensation (Karetsou et al 2004). An up-regulation of this gene has been observed in human gastric cancer (Leys et al 2007) and has been described as a prediction marker for radiosensitivity in rectal cancer (Ojima et al 2007). Phosphodiesterase 6D (PDE6D) interacts with HRAS (Hanzal-Bayer et al 2002, Nancy et al 2002.…”

“…There have been a few studies aimed at predicting response to radiotherapy using gene expression profiling in the treatment of esophageal, rectal, nasopharyngeal cancer and neuroblastoma (4,6,10,11). Identification of the genes differentially expressed between radiosensitive and radioresistant cancers may provide new insights into the mechanisms underlying clinical radioresistance and improve the efficacy of radiotherapy (11).…”
