Zosteran pectin isolated from the perennial marine plant Zostera marina (L.) has a complex molecular structure and a wide range of biological activity. Aim Study: This study aims to identify new properties (adhesive, cryoprotective) of zosteran pectin. Materials and Methods: The adhesive properties of zosteran were investigated by evaluating the microorganism adhesiveness index and the erythrocyte participation rate. To assess the cryoprotective properties of zosteran, the cryoscopic method and various methods for assessing the viability of leukocytes (cryoresistance, cell membrane integrity, and phagocytosis) were used. Results: Zosteran pectin in each assay consistently revealed more adhesive and cryoprotective properties compared to other substances. The results revealed that zosteran at a concentration of 0.3% increase the percentage of red blood cells with lactobacilli attached to their surface. The addition of zosteran to various cryoprotectants changed the osmolarity and freezing point of only the mixture of pectin with DMSO. Zosteran/DMSO cryosolution significantly better retained the viability of leukocytes after exposure to negative temperatures (-80°C), compared with a solution containing only DMSO. Conclusion: Our data reveal new properties of zosteran, due to which pectin changes the conditions of bacterial adhesive activity. This can be used to enhance the perception of microorganisms by intestinal cells in representatives of normal microflora or to reduce it in pathogens of infectious diseases. The obtained cryoprotective effect of the solution with zosteran can be used for further research on the preservation of biological objects (spermatozoa, platelets, etc.) at low temperatures.

In this study, crude extracts of the marine eelgrass Zostera japonica and their solvent-partitioned fractions were evaluated for their inhibitory effect against AGS, HT-1080 and MCF-7 human cancer cells using MTT assay. Each of the crude extracts (acetone/methylene, chloride, and methanol) of Z. japonica showed a significant inhibitory effect on the growth of human cancer cells. The combined crude extracts were partitioned between CH 2 Cl 2 and water. The organic layer was further partitioned between 85% aq. MeOH and n-hexane, and the aqueous layer was then fractionated into n-BuOH and H 2 O, successively. Growth inhibition effects of solvent-partitioned fractions from Z. japonica on human cancer cells increased in a dose-dependent manner. Among these tested samples, the 85% aq. MeOH fraction revealed good inhibitory effects on the growth of AGS and HT-1080 human cancer cells, while the n-hexane fraction exhibited good inhibitory effects on the growth of AGS and MCF-7 human cancer cells. In addition, 85% aq. MeOH and n-hexane fractions enhanced mRNA expression of p53 gene. These results suggest that there is further scope for the isolation of active compounds from Z. japonica, which should show much stronger anticancer activity.

This study aimed to investigate the possible mechanisms of environmental metal pollutant lead (Pb)-induced apoptosis in chicken. Forty 8-day-old healthy chickens were randomly assigned to two groups (n = 20/group) after raising standard commercial diet and drinking water for 1 week: including control group and Pb group ((CHCOO)Pb 350 mg/L of drinking water); the chickens were given euthanasia and collected livers at 90 days. A significant increase of apoptosis rate were found in Pb group and Pb induced obvious ultrastructural changes of chicken liver. The mRNA levels of glycometabolism key enzymes were significantly lower in Pb group than those in controls. Higher levels of malondialdehyde (MDA) and nitric oxide (NO) were observed in Pb group; the activities of antioxidant enzymes and ATPases were significantly lower in Pb group than those in controls, while the inducible nitric oxide synthase (iNOS) activity was on the contrary. The mRNA and protein levels of pro-apoptotic genes were all lower in Pb group than those in controls. Altogether, Pb-induced mitochondrial swelling and nuclear chromatin condensation, oxidative stress, energy metabolism disorder, thereby lead to apoptosis via mitochondrial pathway in chicken liver, suggesting that Pb-induced mitochondrial pathway apoptosis plays an important role in the mechanisms of Pb cytotoxicity to chicken liver.
