Effects of acute ethanol administration and cold exposure on the hypothalamic-pituitary-thyroid axis

Abstract: A single dose of ethanol increases cellular levels of the mRNA encoding thyrotropin-releasing hormone (TRH) in neurons of the paraventricular nucleus (PVN), but blocks the cold-induced increase in the levels of this mRNA. Because the thyrotropic response to cold is dependent upon TRH secretion, we hypothesized that ethanol uncouples the stimulus-induced regulation of TRH secretion from the stimulus-induced regulation of TRH expression. We employed two complementary strategies to test this hypothesis. The first… Show more

“…Furthermore, cold exposure increases TRH mRNA levels within 1 h if the cold stress is initiated in the afternoon, but not in the morning [37]. Other authors were only able to measure a TRH [32] and a TSH [38] response to cold in the afternoon, failed in rats nonacclimatized to elevated ambient temperatures.…”

“…Acute administration of some drugs alters Trh expression and these changes do not always coincide with changes in TSH or TH serum concentrations (Table 1). Although ethanol increases Trh mRNA levels in the PVN, it blocks increases induced by cold-exposure (Zoeller et al 1995). Cold increases Trh mRNA expression independent of high-TH serum levels (Zoeller et al 1990), but a mild stress 2 h before being submitted to cold suppresses the expected increase in Trh mRNA levels in the PVN but not in serum TSH in male rats (Uribe et al 2011).…”

“…The changes in Trh mRNA are transient, with increases at 1 h and normalizing by 2 h, even when animals are kept in the cold for long periods; Trh mRNA levels rise again at 6 h but only if animals are exposed during the light period (Uribe et al 1993, Zoeller et al 1995. Acute administration of some drugs alters Trh expression and these changes do not always coincide with changes in TSH or TH serum concentrations (Table 1).…”

“…In vitro transcription was performed using linearized plasmid and 35 S-UTP (NEN), according to Zoeller et al (1995) with some modifications. Following removal of the template with RNase-free DNase, the RNA was phenol/chloroform extracted, ethanol precipitated, resuspended in RNase-free water to around 100 ng/µl and stored at -80°C.…”
