Effects of cyclohexanone analogues of curcumin on growth, apoptosis and NF-κB activity in PC-3 human prostate cancer cells

Abstract: Abstract. Curcumin is a non-nutritive yellow pigment found in the spice turmeric, which is derived from the rhizome of the plant Curcuma longa Linn. Six cyclohexanone analogues of curcumin (A 1 -A 6 ) were investigated for their effects on growth and apoptosis in PC-3 human prostate cancer cells. The ability of these compounds to inhibit NF-κB activity in PC-3 cells was also determined. Five out of the six curcumin analogues (A 2 -A 6 ) had stronger inhibitory effects compared to curcumin on the growth of cult… Show more

“…It is formerly considered as a drug possessing anti-angiogenic, anti-inflammation and antioxidation effect (Kwon et al, 2005;Suckow et al, 2006;EI-Azab et al, 2011;. Previous researches in 2013 have demonstrated the antitumor effect of Cum in a series of cancers (Bayet-Robert et al, 2013;Du et al, 2013;Kumaravel et al, 2013;Masuelli et al, 2013;Ono et al, 2013;Wei et al, 2013;Yallapu et al, 2013;Zhang et al, 2013). …”

“…Curcumin has the ability to inhibit cell growth and induce apoptosis in both androgen-dependent and androgenindependent prostate cancer cell lines acting through the regulation of apoptosis relative signaling (Hilchie et al, 2010;Wei et al, 2013) and other crucial proteins (Teiten et al, 2012;Horie, 2012;Kang et al, 2013). By targeting the inflammatory cytokines CXCL-1 and -2, curcumin also inhibited prostate cancer metastasis in vivo (Killian et al, 2012).…”

“…The cells were then treated with various concentrations (10~160 μg/ml) of each AT extracts for 72h. To measure cell viability, the MTT assay was performed as described earlier (Wei et al, 2012). The effect of different AT extracts on growth was assessed as percent cell growth as compared to the DMSO-treated cells.…”

“…A minimum of 200 cells were counted to determine the percentage of apoptotic cells in each sample. Caspase-3 activation was measured using an EnzoLyte AMC Caspase-3 Assay Fluorimetric kit (AnaSpec, Fremont, CA, USA) following the manufacturer's instructions (Wei et al, 2012). Fluorescence intensity was measured using a Tecan Infinite M200 plate reader (Tecan US Inc., Durham, NC, USA).…”
