Efficacy of a Chitin-Based Water-Soluble Derivative in Inducing Purpureocillium lilacinum against Nematode Disease (Meloidogyne incognita)

Abstract: Plant-parasitic nematodes cause severe economic losses annually which has been a persistent problem worldwide. As current nematicides are highly toxic, prone to drug resistance, and have poor stability, there is an urgent need to develop safe, efficient, and green strategies. Natural active polysaccharides such as chitin and chitosan with good biocompatibility and biodegradability and inducing plant disease resistance have attracted much attention, but their application is limited due to their poor solubility.… Show more

“…About 10% of nematodes from each treatment were randomly selected to record their head thrashes per minute. 14,17 The dosage effect of 2-furoic acid on the vitality of J2 was identified using a gradient of four concentrations: 25, 50, 75, and 100 μg/mL. The nematodes treated with sterilized water were served as the control group.…”

“…M. incognita was cultured on cucumber, and the nematode eggs were obtained as described previously . To obtain second-stage juveniles (J2s), the isolated eggs were allowed to hatch with sterile distilled water in a Petri dish at 28 °C for 5 days.…”

“…Then, two seeds of two groups were washed with water and placed symmetrically on the edge of a ϕ9 cm petri dish covered with 5 mL of Pluronic F-127 gel (23%) at room temperature, respectively. The gel was prepared according to the method described by Zhan et al [30]. Next, 100 J2s were placed in the center of the dish and incubated in a dark environment for 48 h. The seedlings of the two groups were washed with distilled water and cultured in a humid environment for 7 days.…”

“…Chitin is an insoluble polysaccharide when extracted by alkaline treatment and partially soluble when extracted by enzymatic treatment, from which other compounds of high biological significance are derived, such as chitosan and ChOs [42]. According to Buendia et al [17] and Zhan et al [43], ChOs are soluble depending on the pH and degree of polymerization (dp).…”
