Efficient optimization of ultra‐high‐ performance supercritical fluid chromatographic separation of Rosa sericea by response surface methodology

Abstract: An approach for rapid optimization of ultra-high-performance supercritical fluid chromatographic (UHPSFC) gradient by response surface methodology was developed for fast separation of complex crude extracts of the leaves of Rosa sericea. The optimization was performed with Box-Behnken designs and the multicriteria response variables were described using Derringer's desirability. Based on factorial design experiments, five factors were selected for Box-Behnken designs to optimize the UHPSFC conditions, which le… Show more

“…Box–Behnken design (BBD), a type of RSM, is an independent quadratic design that does not contain an embedded factorial or fractional factorial design . BBD is easy to arrange and interpret in comparison with others . It has been successfully used in the optimization of the extraction of active components such as polyphenols , octyl acetate , aesculin , and ginsenosdes from natural sources.…”

“…The aim of screening column is that all the analytes could be eluted and gets high resolutions with good peak shapes in short time. Although there are some reports about using SFC to separate mixed organic compounds , the theory about how to screen the column according to the sample properties is still developing. There is no detailed guide for choosing the stationary phase in SFC.…”

“…In addition, SFC shows specific selectivity for the similar structure compounds by addition of typical organic modifiers used in reversed‐phase chromatography to CO 2 and diverse range of stationary phases . With the introduction of a new generation of SFC with ultra‐performance convergence chromatography (UPC 2 ), the analysis time and the usage of organic solvent could be drastically reduced . The fast and high‐throughput characteristics of UPC 2 provide the possibility to analyze large amount of water samples in a limited time.…”

“…Recently, ultra-performance convergence chromatography (UPC 2 ), a new generation of supercritical fluid chromatography (SFC), has been developed and shown to have a better separation of non-polar compounds, with reduced analytical time and organic solvent usages (Ruiz-Sambl as et al, 2010;Gourmel et al, 2013;Li et al, 2013). UPC 2 combined with quadrupole time-of-flight mass spectrometry (Q-TOF-MS) has been shown to serve as a powerful tool in investigating triacylglycerol compositions of sunflower, corn and soybean oils by providing accurate mass data and MS 2 fragment ion patterns after improved peak resolution (Zhou et al, 2014;Gao et al, 2017).…”
