Electrochemical impedance spectroscopy for black lipid membranes fused with channel protein supported on solid-state nanopore

Abstract: Black lipid membranes (BLMs) have been used for detecting single-channel activities of pore-forming peptides and ion channels. However, the short lifetimes and poor mechanical stability of suspended bilayers limit their applications in high throughput electrophysiological experiments. In this work, we present a synthetic solid-state nanopore functionalized with BLM fused with channel protein. A nanopore with diameter of ~180 nm was electrochemically fabricated in a thin silicon membrane. Folding and painting t… Show more

“…The thin base layer structures fabricated inside a silicon microstructure had cavity depths of ~493 ± 6 μm was fabricated using 30% KOH at 55 °C for four days followed by room temperature treatment for 4 h. A detailed description for the production of this silicon-based micron cavity was previously discussed in [ 28 ]. Figure 1 shows an image on the left from scanning electron microscopy (SEM) of a typical Si cavity fabricated in this work.…”

“…Many devices of this type exist and are popular on the medical market; the examples are vein filters and catheters used for prevention of pulmonary embolism and drug delivery [48]. Here, we assume that the top of the detector is covered with a layer of channelrhodopsin molecules, which are arranged in a squared matrix and located on a lipid membrane, using so called black lipid membrane technique [49,50]. Above the channelrhodopsin layer ( Fig.…”

“…Promoter expression has been monitored during cell growth, and the variable luciferase activities detected, demonstrating how certain genes are expressed in the gastrointestinal tract [ 66 ]. As an example of the potential of the technique, biosensors could be used to measure antibodies, enzymes, tumor necrosis factor or proteins of interest [ 67 , 68 , 69 , 70 ].…”
