Electron Microscope Studies of the Microvilli of Hela Cells

Abstract: Microvilli of HeLa cells cultured in vitro were preserved for electron microscopic examination at different stages of routine cultivation procedures. By a double-embedding technique, vertical sectioning for electron microscopy was possible. It revealed that, although the microvilli were present on all sides of the cell in the dispersed stage and in the attached stage, they were not present on the bottom of the cell when it was stretched on the surface of the dish. When the cells were grown in dense colonies, t… Show more

“…FIV-infected CRFK cells produced abundant microvilli in the presence or absence of tetherin (Fig. 8A) with a morphology similar to that described previously for HeLa cells (12). While 0.1-m-diameter particles were noted in the regions rich in microvilli, the resolution of the CRFK-derived images made it difficult to distinguish conclusively electrondense virus-like particles from a cross-section through the tip of a microvillus (Fig.…”

“…Oligonucleotide primers were synthesized corresponding to the predicted start (forward primer 5Ј-ATGGCACCTGCTTTTT TACCAC-3Ј) and stop (reverse primer 5Ј-TCAGGCCAGCAGAGCAACGAA-3Ј) codons of feline tetherin. Mya-1 cells were lysed (QIAshredder; Qiagen Ltd., Crawley, United Kingdom), and total RNA was prepared by guanidine-isothiocyanate lysis using an RNeasy Minikit (Qiagen Ltd., Crawley, United Kingdom) and reverse transcribed using a Transcriptor High Fidelity cDNA synthesis kit and oligo(dT) 12 primer (Roche Applied Science, Burgess Hill, United Kingdom). The feline tetherin (feTHN) cDNA was amplified using Phusion DNA polymerase [Finnzymes; New England BioLabs (UK) Ltd., Hitchin, United Kingdom] and its nucleic acid sequence determined using BigDye Terminator v1.1 cycle sequencing (Applied Biosystems, Life Technologies, Paisley, United Kingdom) followed by analysis performed using an Applied Biosystems 3730xl genetic analyzer and chromatogram analysis performed using the Chromas version 1.45 software package (Technelysium Pty.…”

“…One hypothesis is that the cell membrane affinity of sentan is dependent on the curvature of the membrane; as a result, sentan localization to the protrusions could be dependent on their diameter. Interestingly, the diameter of the distal portion of cilia is almost equivalent to the diameter of HeLa cell protrusions: 150 and ϳ120 -130 nm, respectively (Fisher and Cooper, 1967;Porter et al, 1974;Foliguet and Puchelle, 1986;Hosaka et al, 1993). These diameters are smaller than that of cilia axoneme (ϳ250 -300 nm) and larger than that of microvilli (ϳ80 nm) present on the apical surface of tracheal epithelial cells (Simionescu and Simionescu, 1976;Dalen, 1983); neither of these recruited adenovirus-overexpressed or native sentan, which supports the idea that sentan localization is dependent on the curvature of the membrane.…”

“…These results may be interpreted as follows: reversible attachment was caused by long-range forces of the secondary minimum; irreversible attqchment was probably due to the stronger binding forces of the primary minimum and hence must have occurred via the agency of a salmonella adhesin. The calculations were based initially on the assumption that the HeLa cell surfaces were near planar whereas HeLa cells do produce filamentous surface projections (Fisher & Cooper, 1967;Pugh-Humphreys & Sinclair, 1970;Kihlstrom & Latkovic, 1978). Accommodation of this variable and others, however, did not appreciably alter the value of H or the conclusions.…”
