Elevated serum CXCL16 is an independent predictor of poor survival in ovarian cancer and may reflect pro-metastatic ADAM protease activity

Abstract: Background:In certain cancers, expression of CXCL16 and its receptor CXCR6 associate with lymphocyte infiltration, possibly aiding anti-tumour immune response. In other cancers, CXCL16 and CXCR6 associate with pro-metastatic activity. In the current study, we aimed to characterise the role of CXCL16, sCXCL16, and CXCR6 in ovarian cancer (OC).Methods:CXCL16/CXCR6 expression was analysed on tissue microarray containing 306 OC patient samples. Pre-treatment serum sCXCL16 was determined in 118 patients using ELISA… Show more

“…[20][21][22] But, CXCR6 expression is reported to be not associated with lymphocyte infiltration and survival in ovarian cancer. 23 In the present study, CXCR6 was highly expressed in gastric cancer tissues, and was closely correlated with the lymph node metastases in patients with gastric cancer, indicating that CXCR6 might promote the tumorigenesis of gastric cancer. 24 Metastasis is thought to be the clear mark for many aggressive cancers.…”

“…ADAM‐mediated substrate shedding has prognostic potential for ovarian cancer. Soluble CXCL16/CXCR6, in pretreatment serum samples, has been reported to independently predict poor survival for cancer patients (Gooden et al, ), and may therefore serve as a marker that identifies patients with highly metastatic tumors (Gooden et al, ). ADAM17 is a sheddase for activated leukocyte cell adhesion molecule and soluble activated leukocyte cell adhesion molecule [sALCAM] levels were found to be significantly higher in the serum of EOC patients ( P < 0.005) compared to that of healthy controls and correlated with more aggressive Type II tumors (Carbotti et al, ).…”

“…SR-PSOX/CXCL16 and its binding partner CXCR6, as well as its processing enzyme ADAM10 and ADAM17, are overexpressed in many tumor cells, including prostate, breast, colorectal, gastric, liver, ovarian, pancreatic, cervical, lung cancer, glial tumors, and Ewing sarcoma (Gooden et al, 2014; Hattermann, Held-Feindt, Ludwig, & Mentlein, 2013; Huang, Zhang, Cui, Zhao, & Zheng, 2013; Na et al, 2014; Wente et al, 2008). Increasing evidence supports the involvement of sCXCL16 in tumor progression, e.g., promoting proliferation, migration, and invasiveness of CXCR6 + cancer cells (Lu et al, 2008; Schramme et al, 2008; Xing et al, 2012).…”

The elevated level of sCXCL16 in serum has been considered an independent predictor for poor survival in ovarian cancer patients and those with colorectal cancer who have developed the recurrence of liver metastases (Gooden et al, 2014; Matsushita et al, 2012). Independent studies also support the association of SR-PSOX/CXCL16 with poor prognosis in various cancers.…”
