Endocrine and clinical consequences of combination tamoxifen – aminoglutethimide in postmenopausal breast cancer

“…A similar rise in SHBG levels with an associated fall in gonadotrophin levels in patients treated with tamoxifen has been attributed to the partial oestrogen agonist activity of tamoxifen (Sakaiet al, I -J 1978;Willis et al, 1977;Coombes et al, 1982;Dowsett et al, 1984). We have not observed a fall in gonadotrophin levels in zindoxifene treated patients.…”

“…55 The use of HRT 53,56,57 and drugs such as Tamoxifen (adjuvant therapy for breast carcinoma) can also therefore increase the risk of recurrence in the postmenopausal population. [58][59][60][61][62] Postmenopausal endometriosis and HRT Another dilemma in women with endometriosis is the role of HRT in women undergoing radical surgery with bilateral oophorectomies as well as in those going through the natural menopause. As most of the patients undergoing a surgical menopause are young, one needs to carefully consider the benefits of HRT as endometriosis is a hormone-dependent disease and estrogen replacement can be associated with a risk of recurrence or malignant transformation especially in known severe cases and in those that are obese.…”
