It was previously reported that the delta opioid receptor (DOR) agonist SNC80 and antagonist naltrindole modulate the excitability of hippocampal glutamate neurons in primary cultures. The present study aimed to investigate the acute effects of these ligands on the firing activity of hippocampal cornu ammonis 1/3 (CA1/3) glutamate, dorsal raphe nucleus (DRN) serotonin (5-HT), locus coeruleus (LC) noradrenaline, and ventral tegmental area (VTA) dopamine neurons in in vivo conditions. Adult Wistar male rats were used. SNC80 and naltrindole were administered intravenously. Neuronal firing activity was assessed using extracellular single-unit electrophysiology. SNC80, administered first at 1-3 mg/kg, dose-dependently inhibited CA1/3 glutamate, DRN 5-HT, and VTA dopamine neurons. Naltrindole, administered at 1-3 mg/kg after SNC80, did not have any additional effect. Naltrindole, administered first at 1-3 mg/kg, stimulated DRN 5-HT neurons in a dose-dependent manner; this stimulation was dose-dependently reversed by 1-3 mg/kg of SNC80. SNC80 and naltrindole inhibited LC noradrenaline neurons when only they were co-administered at 3 mg/kg, and only when SNC80 was administered first. In conclusion, DOR ligands alter the firing activity of hippocampal glutamate and brainstem monoamine neurons in in vivo conditions. The psychoactive effects of DOR ligands, reported in previous studies, might be explained, at least in part, by their ability to modulate the firing activity of hippocampal glutamate and brainstem monoamine neurons.

Pregnant patients physiologically undergo various changes in their bodies due to hormonal actions. The oral cavity is not exempt from these alterations, and repercussions on oral health are also identified. It is essential to provide safe and appropriate dental care during pregnancy, taking into account the gestational trimester and the patient's overall health. Analgesia in pregnant patients remains a taboo for many professionals, especially for newly graduated dental surgeons. The use of local anesthetics in the office as a pain control method makes the procedure safer and more efficient. Regarding orally administered analgesics, acetaminophen is considered the first choice, with dipyrone as the second option. Aspirin, ibuprofen, and corticosteroids may be an option, but their use should be rationalized and when the benefits outweigh the risks to the maintenance of pregnancy and maternal health. The use of analgesia or reduction of the pain protocol in a pregnant patient in the office can be performed with 2% lidocaine with epinephrine, as it is considered safe. However, certain anesthetics, such as benzocaine and procaine, should be avoided due to risks of methemoglobinemia. Nonetheless, the professional must always weigh the risks and benefits of using analgesic medications for the pregnant patient and the fetus when choosing the medication.

Mice lacking the σ 1 receptor chaperone (σ 1 R −/− ) are resilient to depressive-like behaviors secondary to neuropathic pain. Examining the resilience's brain mechanisms could help develop conceptually novel therapeutic strategies. We explored the diminished motivation for a natural reinforcer (white chocolate) in the partial sciatic nerve ligation (PSNL) model in wild-type (WT) and σ 1 R −/− mice. In the same mice, we performed a comprehensive reverse transcription quantitative PCR (qPCR) analysis across ten brain regions of seven genes implicated in pain regulation and associated affective disorders, such as anxiety and depression. PSNL induced anhedonic-like behavior in WT but not in σ 1 R −/− mice. In WT mice, PSNL up-regulated dopamine transporter (DAT) and its rate-limiting enzyme, tyrosine hydroxylase (Th), in the ventral tegmental area (VTA) and periaqueductal gray (PAG) as well as the serotonin transporters (SERT) and its rate-limiting enzyme tryptophan hydroxylase 2 (Tph2) in VTA. In addition, μ-opioid receptor (MOR) and σ 1 R were up-regulated in PAG, and MOR was also elevated in the somatosensory cortex (SS) but down-regulated in the striatum (STR). Finally, increased BDNF was found in the medial prefrontal cortex (mPFC) and hypothalamus (HPT). Sham surgery also produced PSNL-like expression changes in VTA, HPT, and STR. Genetic deletion of the σ 1 R in mice submitted to PSNL or sham surgery prevented changes in the expression of most of these genes. σ 1 R is critically involved in the supraspinal gene expression changes produced by PSNL and sham surgery. The changes in gene expression observed in WT mice may be related to pain-related depression, and the absence of these changes observed in σ 1 R −/− mice may be related to resilience.
