Endogenous Sulfur Dioxide: A New Member of Gasotransmitter Family in the Cardiovascular System

Abstract: Sulfur dioxide (SO2) was previously regarded as a toxic gas in atmospheric pollutants. But it has been found to be endogenously generated from metabolism of sulfur-containing amino acids in mammals through transamination by aspartate aminotransferase (AAT). SO2 could be produced in cardiovascular tissues catalyzed by its synthase AAT. In recent years, studies revealed that SO2 had physiological effects on the cardiovascular system, including vasorelaxation and cardiac function regulation. In addition, the path… Show more

“…Moreover, these therapeutic molecules, as significant physiological messengers, can maintain the homeostasis of physiological functions in humans . Recently, a number of studies have indicated that sulfur dioxide (SO 2 ) produced by metabolic processes in the body plays a significant role in regulating the cardiovascular system . In addition, elevated concentration of SO 2 could regulate oxidative stress and could cause DNA damage, resulting in the death of cancer cells .…”

“…In addition to H 2 S as the sulfur‐containing gas, sulfur dioxide (SO 2 ) can also be generated in human body via the metabolic process originating from some sulfur‐containing amino acids in the body, such as l ‐cysteine and l ‐methionine. It has also been demonstrated that SO 2 is effective in regulating the cardiovascular system, but the large uptake of SO 2 would cause the severe oxidative damage to organism because of the GSH depletion and ROS generation . Based on the fact that SO 2 gas could enhance the ROS level in tumor cells and then reverse the MDR of cancer cells, Chen et al designed a small molecular prodrug of SO 2 , which was then self‐assembled into micelles with the responsiveness to GSH .…”

“…The endogenously generated sulfur-containing gases H2S and SO2 that are intermediates of the sulfate generating pathways (Fig. 1B), also contribute to numerous physiological and pathophysiological roles in the mammalian body (Medani et al, 2011;Wang et al, 2011;di Masi and Ascenzi, 2013;Wang et al, 2014;Guo et al, 2016;Huang et al, 2016). In particular, both H2S and SO2 modulate vascular tone and cardiac function, and have cardioprotective effects in animal models of systemic and pulmonary hypertension, atherosclerosis and cardiac ischemia-reperfusion injury (Elrod et al, 2007;di Masi and Ascenzi, 2013;Wang et al, 2014).…”
