Endothelial Cell MicroRNA Expression in Human Late-Onset Fuchs' Dystrophy

Abstract: The present study provides the first miRNA profile in FECD and normal endothelial cells and demonstrates widespread miRNA downregulation in FECD. Decreased endothelial expression of miR-29 family members may be associated with increased subendothelial extracellular matrix accumulation in FECD.

“…However, in many patients the inflammation does not resolve completely, and this culminates in corneal opacification and loss of vision. 38 Recent reports on altered miRNA expression in human corneal diseases [10][11][12] suggest their regulatory role in pathogenesis. However, the miRNA expression profile in fungal keratitis has not yet been studied.…”

“…7 MicroRNAs are expressed in various ocular tissues with a distinct tissue-specific and developmental stage-specific expression pattern, which suggests potential unique functions. 8 They have also been detected in ocular fluids 9 and have been implicated in human corneal diseases like keratoconus, 10 Fuchs' dystrophy, 11 and herpetic stromal keratitis. 12 Corneal inflammation in fungal keratitis poses a significant therapeutic challenge to clinicians, particularly because of the controversial role of corticosteroids in the initial disease management.…”

“…67 Evidence of downregulation of three miR-29 family members (miR-29a-3p, miR-29b-2-5p, and miR-29c-5p) is further supported by upregulation of collagen I and collagen IV, mRNA targets which are associated with extracellular matrix production, and increased endothelial laminin protein expression FCD. 67 …”
